Show moreUrbanization has become a global trend under the impact of population growth, socio-economic development, and globalization. However, the interactions between climate change and urban growth in the context of economic geography are unclear due to missing links in between the recent planning megacities. This study aims to conduct a multi-temporal change analysis of land use and land cover in New York City, City of London, and Beijing using a cellular automata-based Markov chain model collaborating with fuzzy set theory and multi-criteria evaluation to predict the city's future land use changes for 2030 and 2050 under the background of climate change.To determine future natural forcing impacts on land use in these megacities, the study highlighted the need for integrating spatiotemporal modeling analyses, such as Statistical Downscale Modeling (SDSM) driven by climate change, and geospatial intelligence techniques, such as remote sensing and geographical information system, in support of urban growth assessment. These SDSM findings along with current land use policies and socio-economic impact were included as either factors or constraints in a cellular automata-based Markov Chain model to simulate and predict land use changes in megacities for 2030 and 2050. Urban expansion is expected in these megacities given the assumption of stationarity in urban growth process, although climate change impacts the land use changes and management. More land use protection should be addressed in order to alleviate the impact of climate change.

Granuloma and pathologic manifestations in cd resemble aspects found in tuberculosis, leprosy and paratuberculosis. The latter, a chronic enteritis in cattle, goat, sheep and primates, which is similar to human enteritis, also known as cd, is caused by a fastidious, slow growing mycobacterium avium subspecies paratuberculosis (map). Due to the similarities between cd and paratuberculosis, a mycobacterial cause in cd has been proposed. Recent discovery of a possible association between nod2/card15 mutations and risk of cd added support to microorganism-host interactions. In this study, a possible mycobacterial role in cd etiology has been evaluated by investigating the presence of map dna, the state of the cellular immune response and microarray gene expression profiling in peripheral blood and surgical tissue from cd, uc and healthy control subjects. Nested pcr detected map dna in tissue from 10/12(83%) cd patients compared to 1/6(17%) non-ibd subjects. Fluorescence in situ hybridization (fish) with the aid of confocal scanning laser microscopy (cslm) detected map dna in 8/12(67%) cd subjects compared to 0/6(0%) in non-ibd subjects. The detection of map dna by either technique in tissue from cd subjects is significant compared to non-ibd subjects (p < 0.05). Map dna was also detected in both inflamed and non-inflamed tissue from patients with cd suggesting map infiltration in human tissue. Correlation of possible map presence and the function of polymorphonuclear leukocytes (pmn) and peripheral blood mononuclear cells (pbmc) in 19 cd patients and 12 controls have been evaluated. Pmn phagocytosis of viable fitc-map was suppressed in 13/19(68%) cd patients compared to 0/12(0%) in healthy controls (p<0.05). Pbmc phagocytosis of viable fitc-map was suppressed in 5/19(26%) of cd patients compared to 0/12(0%) of healthy controls (p<0.05). The proliferative response of pbmc with t-cell majority from cd and controls subjects was evaluated against pha, candida albicans, pwm and map ppd. Dysfunctional proliferative response against pha was found in 8/19(42%) cd patients compared to 1/12(8.3%) in controls suggesting possible t-cell anergy. Pbmc from 11 cd subjects reacted normally to pha, 7/11(64%) reacted strongly to map ppd suggesting previous exposure to mycobacteria, and 3/11(27%) did not react with map ppd suggesting lack of pre-exposure to mycobacteria. From the seven mycobacterial pre-exposed samples, 6/7(86%) showed a normal ability to recall antigens by activated macrophages when exposed to c. Albicans, and all 7 samples had a normal pwm response. Finally, microarray-chip technology was employed to identify the expression profile of genes that have a role in the immune response of cd patients. Rna was isolated from fresh buffy coats from 8 healthy controls, 2 cd, and 1 uc patients. Chips with an estimated of 30,000 human genes were hybridized to cdna from these samples. We found that 17% of the total number of genes was differentially expressed. Over 200 genes were involved in the immune response, 7 genes where common to both forms of ibd (uc and cd), and 8 genes were found to be either downregulated in cd and upregulated in uc or viceversa. The ifngr1 gene, which encodes the ligand-binding chain of the ifn-gamma receptor, was found to be downregulated in 2/2(100%) of cd patients, but not in uc patients. It is known that defects in ifngr1 are a cause of atypical mycobacterial infection and bcg infection. Patients suffering from this deficiency have an immunologic defect predisposing them to infection with mycobacteria. This correlates with the proposed theory as map being the causative agent of cd. Furthermore, the results indicate a host susceptibility requirement for the establishment of mycobacterial infection in cd patients. Further characterization of ifngr1 using real-time pcr is underway. Collectively, detection of map dna in the majority of cd tissue and the alteration in pmn and pbmc to respond efficiently to map may be related to the fact that mycobacterial pathogens infect phagocytic cells of susceptible hosts and consequently the immune response is dysregulated. Furthermore, the fact that a gene linked to mycobacterial susceptibility was found to be downregulated in cd patients only, strengthens the mycobacterial etiology of cd. In general, the data suggest a possible role for a bacterial pathogen in cd pathogenesis.

To satisfy users' expectation from wireless data services, researchers and practitioners recognized the necessityof more utilization and pervasive sharing of the spectrum. Though scarce, spectrum is underutilized in some areas or within certain operating hours due to the lack of appropriate regulatory policies, static allocation and emerging business challenges. Thus, finding ways to improve the utilization of this resource to make sharing more pervasive is of great importance. There already exists a number of solutions to increase spectrum utilization via increased sharing. Dynamic Spectrum Access (DSA) enables a cellular operator to participate in spectrum sharing in many ways, such as geological database and cognitive radios, but these systems perform spectrum sharing at the secondary level (i.e., the bands are shared if and only if the primary/licensed user is idle) and it is questionable if they will be sufficient to meet the future expectations of the spectral efficiency.Along with the secondary sharing, spectrum sharing among primary users is emerging as a new domain of future mode of pervasive sharing. We call this type of spectrum sharing among primaryusers as (")pervasive spectrum sharing (PSS)("). However, such spectrum sharing among primary users requires strong incentives to share and ensuring a freeriding-free cellular market.Freeriding in pervasively shared spectrum markets (be it via government subsidies/regulations or self-motivated coalitions among cellular operators) is a real techno-economic challenge to be addressed. In a PSS market, operators will share their resources with primary users of other operators and may sometimes have to block their own primary users in order to attain sharing goals. Small operators with lower quality service may freeride on large operators' infrastructure in such pervasively shared markets. Even worse, since small operators' users may perceive higher-than-expected service quality for a lower fee, this can cause customer loss to the large operators and motivate small operators to continue freeriding with additional earnings from the stolen customers. Thus, freeriding can drive a shared spectrum market to an unhealthy and unstable equilibrium. In this work, we model the freeriding by small operators in shared spectrum markets via a game-theoretic framework. We focus on a performance-based government incentivize scheme and aim to minimize the freeriding issue emerging in such PSS markets. We present insights from the model and discuss policy and regulatory challenges.

Show moreThe single largest cause of compound attrition during drug development is due to inadequate tools capable of predicting and identifying protein interactions. Several tools have been developed to explore how a compound interferes with specific pathways. However, these tools lack the potential to chronically monitor the time dependent temporal changes in complex biochemical networks, thus limiting our ability to identify possible secondary signaling pathways that could lead to potential toxicity. To overcome this, we have developed an in silico neuronal-metabolic model by coupling the membrane electrical activity to intracellular biochemical pathways that would enable us to perform non-invasive temporal proteomics. This model is capable of predicting and correlating the changes in cellular signaling, metabolic networks and action potential responses to metabolic perturbation.The neuronal-metabolic model was experimentally validated by performing biochemical and electrophysiological measurements on NG108-15 cells followed by testing its prediction capabilities for pathway analysis. The model accurately predicted the changes in neuronal action potentials and the changes in intracellular biochemical pathways when exposed to metabolic perturbations. NG108-15 cells showed a large effect upon exposure to 2DG compared to cyanide and malonate as these cells have elevated glycolysis. A combinational treatment of 2DG, cyanide and malonate had a much higher and faster effect on the cells. A time-dependent change in neuronal action potentials occurred based on the inhibited pathway. We conclude that the experimentally validated in silico model accurately predicts the changes in neuronal action potential shapes and proteins activities to perturbations, and would be a powerful tool for performing proteomics facilitating drug discovery by using action potential peak shape analysis to determine pathway perturbation from an administered compound.

Due to the high resilience, relatively low costs, and advanced features of modern hand-held communication devices, these devices are in a position to drastically improve communication flow during emergency management situations. Due to the lack of official implementation of these devices and the lack of the establishment of standard guidelines for device selection, the use of hand-held communication devices in emergency management is yet to be optimized. Island nations such as the Bahamas, which face unique challenges in regard to emergency management due to geographical, infrastructural, political, and cultural hurdles which are found in the region, can especially benefit from the optimized implementation of hand-held communication devices in emergency management. This study examined current emergency response procedures in The Bahamas, created a baseline for the current use of hand-held communication devices by Bahamian emergency management officials and civilians, identified the communication needs of Bahamian emergency management officials and civilians, and proposed a model for the selection of hand-held communication devices based upon human factors principals and focusing on user priorities. This study began with a focus group interview which included 14 Bahamian emergency management officials in order to gain an understanding of current Bahamian emergency response procedures and the communication challenges faced by emergency management officials during high consequence emergencies. A paper based survey was conducted, in which 31 Bahamian emergency management officials answered demographic, skill level, and functionality questions related to the use of hand-held communication devices to support emergency related activities including those directed toward preparation, mitigation, and response. These emergency management officials provided invaluable input based upon their practical experience in high consequence emergency situations. 155 Bahamian civilians participated in a similar survey which was a reduced version of the survey used for emergency management officials. Both surveys included questions in regard to the background information of the participants, previous handheld communication experience, device performance, and what other communication devices were being utilized. The surveys were analyzed using statistical methods of categorical data analysis and correlations were identified. Several communication needs which were categorized as infrastructure, organizational, and equipment needs as well as a hierarchy of device selection factors in regard to the use of hand-held communication devices during emergency management situations were identified. The analytic hierarchy process (AHP) was used in order to determine the priorities of each of the identified device selection factors and a model for the selection of hand-held communication devices used to support communication flow in high consequence emergency management was proposed.

One of the major problems at signalized intersection is the dilemma zone, which is caused by false driver behavior during the yellow interval. This research evaluated driver behavior during the yellow interval at signalized intersections and compared different dilemma zone countermeasures. The study was conducted through four stages.First, the driver behavior during the yellow interval were collected and analyzed. Eight variables, which are related to risky situations, are considered. The impact factors of drivers' stop/go decisions and the presence of the red-light running (RLR) violations were also analyzed. Second, based on the field data, a logistic model, which is a function of speed, distance to the stop line and the lead/follow position of the vehicle, was developed to predict drivers' stop/go decisions. Meanwhile, Cellular Automata (CA) models for the movement at the signalized intersection were developed. In this study, four different simulation scenarios were established, including the typical intersection signal, signal with flashing green phases, the intersection with pavement marking upstream of the approach, and the intersection with a new countermeasure: adding an auxiliary flashing indication next to the pavement marking. When vehicles are approaching the intersection with a speed lower than the speed limit of the intersection approach, the auxiliary flashing yellow indication will begin flashing before the yellow phase. If the vehicle that has not passed the pavement marking before the onset of the auxiliary flashing yellow indication and can see the flashing indication, the driver should choose to stop during the yellow interval. Otherwise, the driver should choose to go at the yellow duration. The CA model was employed to simulate the traffic flow, and the logistic model was applied as the stop/go decision rule. Dilemma situations that lead to rear-end crash risks and potential RLR risks were used to evaluate the different scenarios. According to the simulation results, the mean and standard deviation of the speed of the traffic flow play a significant role in rear-end crash risk situations, where a lower speed and standard deviation could lead to less rear-end risk situations at the same intersection. High difference in speed are more prone to cause rear-end crashes. With Respect to the RLR violations, the RLR risk analysis showed that the mean speed of the leading vehicle has important influence on the RLR risk in the typical intersection simulation scenarios as well as intersections with the flashing green phases' simulation scenario.Moreover, the findings indicated that the flashing green could not effectively reduce the risk probabilities. The pavement marking countermeasure had positive effects on reducing the risk probabilities if a platoon's mean speed was not under the speed used for designing the pavement marking. Otherwise, the risk probabilities for the intersection would not be reduced because of the increase in the RLR rate. The simulation results showed that the scenario with the pavement marking and an auxiliary indication countermeasure, which adds a flashing indication next to the pavement marking, had less risky situations than the other scenarios with the same speed distribution. These findings suggested the effectiveness of the pavement marking and an auxiliary indication countermeasure to reduce both rear-end collisions and RLR violations than other countermeasures.

These non-kinetic operations include rebuilding indigenous institutions' bottom-up economic activity and constructing necessary infrastructure since the success in non-kinetic operations depends on understanding and using social and cultural landscape. This study aims to support decision makers by building a computational model to understand economic factors and their effect on adverse events.In this dissertation, the analysis demonstrates that the use of cellular automata has several significant contributions to support decision makers allocating development funds to stabilize regions with higher adverse event risks, and to better understand the complex socio-economic interactions with adverse events. Thus, this analysis was performed on a set of spatial data representing factors from social and economic data. In studying behavior using cellular automata, cells in the same neighborhood synchronously interact with each other to determine their next states, and small changes in iteration may yield to complex formations of adverse event risk after several iterations of time. The modeling methodology of cellular automata for social and economic analysis in this research was designed in two major implementation levels as follows: macro and micro-level. In the macro-level, the modeling framework integrates population, social, and economic sub-systems. The macro-level allows the model to use regionalized representations, while the micro-level analyses help to understand why the events have occurred. Macro-level subsystems support cellular automata rules to generate accurate predictions. Prediction capability of cellular automata is used to model the micro-level interactions between individual actors, which are represented by adverse events.The results of this dissertation demonstrate that cellular automata model is capable of evaluating socio-economic influences that result in changes in adverse events and identify location, time and impact of these events. Secondly, this research indicates that the socio-economic influences have different levels of impact on adverse events, defined by the number of people killed, wounded or hijacked. Thirdly, this research shows that the socio-economic, influences and adverse events that occurred in a given district have impacts on adverse events that occur in neighboring districts. The cellular automata modeling approach can be used to enhance the capability to understand and use human, social and behavioral factors by generating what-if scenarios to determine the impact of different infrastructure development projects to predict adverse events. Lastly, adverse events that could occur in upcoming years can be predicted to allow decision makers to deter these events or plan accordingly if these events do occur.
