Show moreThe overall focus of this research project was to develop an in vitro tissue-engineered system that accurately reproduced the physiology of the sensory elements of the stretch reflex arc as well as engineer the myelination of neurons in the systems. In order to achieve this goal we hypothesized that myelinating culture systems, intrafusal muscle fibers and the sensory circuit of the stretch reflex arc could be bioengineered using serum-free medium formulations, growth substrate interface design and microfabrication technology. The monosynaptic stretch reflex arc is formed by a direct synapse between motoneurons and sensory neurons and is one of the fundamental circuits involved in motor control. The circuit serves as a proprioceptive feedback system, relaying information about muscle length and stretch to the central nervous system (CNS). It is composed of four elements, which are split into two circuits. The efferent or motor circuit is composed of an α-motoneuron and the extrafusal skeletal muscle fibers it innervates, while the afferent or sensory circuit is composed of a Ia sensory neuron and a muscle spindle. Structurally, the two muscular units are aligned in parallel, which plays a critical role modulating the system's performance. Functionally, the circuit acts to maintain appropriate muscle length during activities as diverse as eye movement, respiration, locomotion, fine motor control and posture maintenance. Myelination of the axons of the neuronal system is a vertebrate adaptation that enables rapid conduction of action potentials without a commensurate increase in axon diameter. In vitro neuronal systems that reproduce these effects would provide a unique modality to study factors influencing sensory neuronal deficits, neuropathic pain, myelination and diseases associated with myelination. In this dissertation, results for defined in vitro culture conditions resulting in myelination of motoneurons by Schwann cells, pattern controlled myelination of sensory neurons, intrafusal fiber formation, patterned assembly of the mechanosensory complex and integration of the complex on bio-MEMS cantilever devices. Using these systems the stretch sensitive sodium channel BNaC1 and the structural protein PICK1 localized at the sensory neuron terminals associated with the intrafusal fibers was identified as well as the Ca2+ waves associated with sensory neuron electrical activity upon intrafusal fiber stretch on MEMS cantilevers. The knowledge gained through these multi-disciplinary approaches could lead to insights for spasticity inducing diseases like Parkinson's, demyelinating diseases and spinal cord injury repair. These engineered systems also have application in high-throughput drug discovery. Furthermore, the use of biomechanical systems could lead to improved fine motor control for tissue-engineered prosthetic devices.

Show moreVertical structure, the top-to-bottom arrangement of aboveground vegetation, is an important component of forest and shrubland ecosystems. For many decades, ecologists have used foliage height profiles and other measures of vertical structure to identify discrete stages in post-disturbance succession and to quantify the heterogeneity of vegetation. Such studies have, however, required resource-intensive field surveys and have been limited to relatively small spatial extents (e.g., <15 ha). Light detection and ranging (lidar) is an active remote sensing technology with enormous potential to characterize the three-dimensional structure of vegetation over broad spatial scales. In this study, discrete-return lidar data were used to create vertical profiles for over 500 vegetation patches on approximately 1000 ha of an oak scrub landscape in the Kennedy Space Center/Merritt Island National Wildlife Refuge area on the east-central coast of Florida. Nonparametric multivariate analysis of variance (NPMANOVA) tests detected significant differences among the profiles belonging to the four predominant land use/land cover (LULC) types in the study area. For the dominant LULC category (Herbaceous upland non-forested), pairwise NPMANOVA comparisons indicated that there were significant differences between vertical profiles for some of the distinct time since fire (TSF) values. Measures of vertical structural diversity (VSD) were calculated from the vertical profiles and then null, linear, and quadratic models relating VSD to TSF were compared via an Akaike information criterion (AIC) model selection procedure. As predicted by the Intermediate Disturbance Hypothesis, the quadratic model was the best model for the Herbaceous upland non-forested LULC category, but it explained less than 3% of the total variation in VSD. When fire frequency was considered in conjunction with TSF for this LULC category, however, the model that was quadratic in both predictor variables was the best model among the candidates and explained over 6% of the total variation in VSD. These results support the Extended Keystone Hypothesis, which predicts that disturbance generates discrete structural patterns across landscapes, and the Intermediate Disturbance Hypothesis, since the VSD of the predominant LULC category was maximized at intermediate levels of fire disturbance (i.e., intermediate values of TSF and/or fire frequency). In addition to demonstrating the ability of discrete-return lidar to characterize the vertical structure of vegetation at the landscape scale, this research has potential management implications. Using the techniques developed in this study, practitioners can compare the vertical structure of managed ecosystems to reference natural systems to evaluate the efficacy of managed disturbance regimes.

This study examines Peace Corps work in Colombia during the 1960s within the framework of U.S. Cold War policy. It explores the experiences of volunteers in Colombia and contrasts their accounts with Peace Corps reports and presentations to Congress. It intends to show the agency's assessment of volunteer work and how it compares to the volunteers' views and Congressional reports. Although the Peace Corps presented some topics and themes expressed by volunteers, the thesis exposes the discrepancies that existed between Peace Corps reports and the volunteers' experiences. Volunteer accounts reveal that there were some criticisms and stories that the agency did not report. Furthermore, evidence sheds light on the obstacles volunteers encountered, how they were presented by the Peace Corps, as well as the value of volunteer work as perceived by volunteers. Finally, the Peace Corps articulated a goal of making friends in the underdeveloped world, and the accounts of the volunteers support the Peace Corps assertion that volunteers were successful in fostering relations and understanding in Colombia during the 1960s.

This thesis was undertaken due to the significant gap in work done by aca-fan (-) a portmanteau of academic and fan(-) scholars who have strong links to the fanfiction community and culture. The aspects explored are the technical writing skills and techniques demonstrated in fanfiction reviews, the influence of the nontraditional online learning environment, the rhetorical strategies that reviewers use to give feedback, the significant categories of things that reviewers comment on, and the value of skills taught peer-to-peer in this manner. The results of my research suggest that peer review in a relaxed, non-academic context leads to improved confidence and skill among a wide demographic range. This thesis proposes that fanfiction writing, reading, and reviewing supports learning. The evidence suggests that it be incorporated where applicable in formal classroom learning to supplement traditional understandings of grammar, syntax, tone, and the use of universal tropes.

This study employs a qualitative, grounded theory approach to examine how sexual minority youth and their allies navigate gender, sexuality, and social movement participation. A year and a half of observation and 16 semi-structured individual interviews were conducted with Gay/Straight Alliance members in a high school setting in the southeastern United States. The study reveals that, through the lens of frame analysis, the G/SA is analogous to larger and more organized social movement organizations. The findings also suggest members often struggle and engage with issues surrounding sexuality, including its origins, coming out as a process, and judgments and evaluations surrounding sex and desire. Additionally, the findings address elements of gender conformity and non-conformity.

Show moreNational and international research on governmental privatization efforts reflects myriad successes and failures. However, little is known about the effectiveness of family safety privatization efforts in the state of Florida. In Brevard County, Florida, family safety privatization efforts have been underway for several years now, and while evaluations are taking place, they do not reflect one key piece of information(-)the perceptions of family safety workers. A snowball sample was obtained from former and current child safety workers and open- and closed-ended questions were administered with a total of 15 former and current family safety workers who work or worked for several different public and private family safety agencies within Brevard County, Florida. Information was obtained regarding perceptions of privatization to adequately and more efficiently do the work of public entities. The results show two primary areas of interest. The model of care which has been instituted post-privatization (CARES) has been perceived as more effective than the former state model; the strongest problematic themes that developed concerned power, control, and the perception of unfairness. These themes are explored using a backwards mapping approach and recommendations for continued growth and cohesion are explored.

Show moreThis project focuses on persistent gaps in philosophies of the body: the enduring mind-body divide in accounts of phenomenology, the unfulfilled promises of representing and inhabiting the body in online and virtual spaces, and the difference between health as quantified in medical discourse versus health as lived experience. These tensions are brought to light through the electronic food journal genre where the difficulty in capturing pre-noetic, outside-consciousness aspects of experience and embodied health are thrown into relief against circulating cultural discourses surrounding health, body size, self-surveillance, and self-care. The electronic food journal genre serves as a space for users to situate themselves and their daily practices in relation to medicalization, public policy, and the conflation of health and body size. These journals form artifacts reflecting life writing practices in digital spaces that model compliant self-surveillance as well as transgressive self-care. The journals instantiate the mind-body-technology interactivity of extended cognition, but also point toward a rupture in the feedback loops that promise to integrate pre-noetic aspects of being and experience. By exploring the tensions inherent in these online food journaling spaces, this project concludes by offering a PEERS heuristic/heuretic for assessing theories and technologies of embodiment and health for their ability to access what resides in the (")remainder(") of current embodiment philosophy and to identify the aspects of lived experience left unattended in USDA health policy, food journaling interfaces, and embodiment philosophy. The PEERS model can be used to evaluate existing technologies for their capacity to map true mind-body-technology interactivity and to build new theory that accounts for a fuller, more nuanced approach to understanding embodied reality and embodied health.

Show moreFor more than a half a century the role of education and its influence on social mobility and status attainment has been a subject of research. Further more, education has been shown to be an important contributor for success over the life course. Much of the research surrounding status attainment and higher education has dealt with the Baby Boomer cohort. The purpose of the study is to examine education from a perspective that is less talked about to this point. This study uses data gathered by the Pew Research Center and examines a specific age group, current 18-30 year olds (Millennials), to gather a better understanding of their attitudes towards the value of higher education within the current era of the economy, education, and job opportunity. According to the analysis, females report higher odds of feeling that a college degree is important to success later in life. Equally important, results indicate that Blacks have greater odds of perceiving education to be important for success in life. Results also demonstrated that in this particular study, other factors such as income and employment status did not significantly affect respondent's perceptions on the importance of education.

Show moreIn sports discourse, the relationship between athletics and technology is often paradoxical. On the one hand, modern sports rely on technology at every level, from training and tracking of players to the equipment and apparel used by athletes to the game strategies and playing fields themselves. Nearly all of these technologies are intended to increase athletic performance on some level. And yet, certain performance enhancement technologies can be criticized for being antithetical to the spirit of sports, which is framed as being a strictly natural and pure human endeavor. Using a rhetorical-cultural methodological approach, popular sports discourse is analyzed to investigate how arguments in contested spaces between sports and technologies get (re)negotiated and (re)articulated to fit within a sports social language that emphasizes (")pure(") and (")natural(") ideals of sport. This often results in a dichotomy where the sport/technology relationship is either black boxed, thus being subsumed in the sport social language and becoming transparent and the relationships unarticulated, or the technology is regulated out of the sport through rules and bans. The reason for this articulation is attributed in large part to the deep humanism embedded in the sport social language. How a shift to a posthuman perspective would effect sports discourse is explored. These conclusions about underlying values in sports discourse lead to the formation of a new theoretical framework called disciplinary mythologies. Building off of Foucault's disciplinary power, Scott's disciplinary rhetorics, and Barthe's mythologies, disciplinary mythologies are discrete units of persuasion that both construct and constitute claims by drawing upon layered narratives and shifting associations that lose their context when entering the realm of myth. Two specific disciplinary mythologies are discussed(-)the level-playing-field topos and the nostalgia enthymeme(-)and it is shown how sports discourse often draws upon them to shape arguments and actions.

I analyze literature from within technical communication to determine how narrative is perceived, as well as the traditions in technical communication that made the field resistant to the use of narrative in the past. These findings are considered with respect to the history and definition of narrative, as well as to how narrative can improve learning outcomes when compared to expository learning approaches commonly used in technical documentation.While narrative is not a new concept to technical communication, this thesis offers new insights through a multidisciplinary approach that considers the work of philosophers and narratologists that are relatively unknown to the field. Philosophers Daniel Dennett and Jerome Bruner, as well as narratologists Gerard Genette, David Rudrum, and David Darby, show that narrative forms the basis for the construction of reality and that all human learning is based on the stories that we construct to give meaning to the world. Research studies conducted on the efficacy of narrative based learning are discussed in detail and an analysis of the areas where narrative use would most benefit technical communication is provided. Recommendations are made for the future use of narrative in technical documentation and for further research on the implementation and cost of narrative solutions.

Show moreThis work describes theoretical advances in the modeling and simulation of microfluidic systems and demonstrates the practical application of those techniques. A new multi-scale model of the adsorption of hard spheres was formulated to bridge the gap between simulations of discrete particles and continuum fluid dynamics. A whispering gallery mode (WGM) biosensor was constructed and used to measure the kinetics of adsorption for two types of proteins on four different surfaces. Computational fluid dynamics was used to analyze the transport of proteins in the flow cell of the biosensor. Kinetic models of protein adsorption that take transport limitations into account were fitted to the experimental data and used to draw conclusions about the mechanisms of adsorption. Transport simulations were then applied to the practical problem of optimizing the design of a microfluidic bioreactor to enable (")plugs(") of fluid to flow from one chamber to the next with minimal dispersion. Experiments were used to validate the transport simulations. The combination of quantitative modeling and simulation and experiments led to results that could not have been achieved using either approach by itself. Simulation tools that accurately predict transport and protein adsorption will enable the rational design of microfluidic devices for biomedical applications.

Show moreThe single largest cause of compound attrition during drug development is due to inadequate tools capable of predicting and identifying protein interactions. Several tools have been developed to explore how a compound interferes with specific pathways. However, these tools lack the potential to chronically monitor the time dependent temporal changes in complex biochemical networks, thus limiting our ability to identify possible secondary signaling pathways that could lead to potential toxicity. To overcome this, we have developed an in silico neuronal-metabolic model by coupling the membrane electrical activity to intracellular biochemical pathways that would enable us to perform non-invasive temporal proteomics. This model is capable of predicting and correlating the changes in cellular signaling, metabolic networks and action potential responses to metabolic perturbation.The neuronal-metabolic model was experimentally validated by performing biochemical and electrophysiological measurements on NG108-15 cells followed by testing its prediction capabilities for pathway analysis. The model accurately predicted the changes in neuronal action potentials and the changes in intracellular biochemical pathways when exposed to metabolic perturbations. NG108-15 cells showed a large effect upon exposure to 2DG compared to cyanide and malonate as these cells have elevated glycolysis. A combinational treatment of 2DG, cyanide and malonate had a much higher and faster effect on the cells. A time-dependent change in neuronal action potentials occurred based on the inhibited pathway. We conclude that the experimentally validated in silico model accurately predicts the changes in neuronal action potential shapes and proteins activities to perturbations, and would be a powerful tool for performing proteomics facilitating drug discovery by using action potential peak shape analysis to determine pathway perturbation from an administered compound.

At the county-level, Iowa and Virginia are analyzed using crime data from the National Incident Based Reporting System. More specifically, gun availability is tested as a mediator for county drug crime rates and homicide counts. Variable selection and prediction is based on routine activity and social disorganization theories. I argue that social disorganization allows the context for which criminal opportunity presents itself through routine activities. I posit gun availability mediates a positive relationship between illegal drug markets and homicide, with differences between urban and rural communities.

Show moreMigration is a widespread and complex phenomenon in nature that has fascinated humans for centuries. Connectivity among populations influences their demographics, genetic structure and response to environmental change. Here, I used the loggerhead turtle (Caretta caretta, L.) as a study organism to address questions related to migratory connectivity and carry-over effects using satellite telemetry, stable isotope analysis and GIS interpolation methods. Telemetry identified foraging areas previously overlooked for loggerheads nesting in Florida. Next, I validated and evaluated the efficacy of intrinsic markers as a complementary and low cost tool to assign loggerhead foraging regions in the Northwest Atlantic Ocean (NWA), using both a spatially implicit and spatially explicit (isoscapes) approach. I then focused on the nesting beaches and developed a common currency for isotopic studies based on unhatched eggs, which provide a non-invasive and non-destructive method for more extensive sampling to elucidate isotopic patterns across broader spatiotemporal scales. Lastly, I found that intra-population variations in foraging strategies affect annual and long-term reproductive output of loggerheads nesting in Florida. Understanding geospatial linkages is critical to the fostering of appropriate management and conservation strategies for migratory species. My multi-faceted approach contributes to the growing body of literature exploring migratory connectivity and carry-over effects.
