Show moreUniversity of Central Florida College of Education Thesis; The purpose of the study was to design a simulation using apersonal computer-based authoring system that could serve as a vehicle for investigating decision-making styles of educational administrators. The program was designed to be used at the graduate level for students seeking advanced degrees in educational leadership. The simulation makes use of simulation techniques for controlling presentation of material and data collection for subequent analysis about information search and utilization procedures of administrators. The study was undertaken to demonstrate that nontechnical personnel using state-of-the-art technology could develop affordable and effective multimedia simulations. The study was also designed to demonstrate that technology was sufficiently advanced that time to develop such a program would not preclude its use. Data were gathered from seven graduate students attending a introductory graduate course. Other sources of data included practicing unitersity professors, district administrators practicing principles. Data were gathered using a survey and personal interviews. Results of the survey indicated that respondents found that the technical approach was innovative and could be useful in teaching graduate students how to be better principals. District administrators expressed interest in using the program to evaluate candidates for beginning principalships.

Show moreRelationships between organic molecules and inorganic minerals are investigated in five carbonaceous chondrites, Northwest Africa 852 (CR2), Tagish Lake (C2-ungroupped), Orgueil (CI1), Sutter's Mill (CM), and Murchison (CM2), with micron spatial resolution using synchrotron-based imaging micro-FTIR spectroscopy. Correlations based on absorption strength for various constituents are determined using statistical correlation analysis. Silicate band is found to be positively correlated with stretching modes of aliphatic hydrocarbons in NWA 852 and Tagish Lake. The former is highly correlated with the hydration band in all meteorites. Negative correlation is observed between water+organics and carbonate bands in all meteorites. Two dimensional infrared maps for NWA 852 and Orgueil show that carbonates are spatially separated from water+organic combination, silicates, OH, and CH distributions. Overlapping of the latter three in NWA 852 and Tagish Lake suggests a possible catalytic role of phyllosilicates in the formation of organics. Additionally, spectroscopic analyses on Sutter's Mill meteorite fragments present multiple distinct mineralogies. Spatial and spectral evidences on this regolith breccia suggest mixing of multiple parent bodies. Ratios of asymmetric CH2 and CH3 band strengths for NWA 852, Tagish Lake, and Sutter's Mill are similar to the average ratio of interplanetary dust particles and Wild 2 cometary dust particles, however significantly exceeds that of interstellar medium objects and several aqueously altered carbonaceous chondrites such as Orgueil. This suggests distinct formation regions and/or parent body processing of organics for these meteorites. Our infrared spectro-microtomography measurements on Murchison meteorite, representing the first such measurement on any kind of meteorite, comprise of three-dimensional reconstructions of specific molecular functional groups for understanding the spatial distributions of these groups.

The growth of high-quality MoS2 by a scalable and doping compatible method is still lacking. Therefore, the suitable dopants for MoS2 are not fully explored yet. This dissertation consists mainly of four main studies. The first study is on the growth of MoS2 thin films by atmospheric pressure chemical vapor deposition. Scanning electron microscope images revealed the growth of microdomes of MoS2 on top of a smooth MoS2 film. These microdomes are very promising as a broadband omnidirectional light trap for light harvesting applications. The second study is on the growth of MoS2 thin films by low pressure chemical vapor deposition (LPCVD). Control of sulfur vapor flow is essential for the growth of a pure phase of MoS2. Turning off sulfur vapor flow during the cooling cycle at 700 (&)#186;C leads to the growth of highly textured MoS2 with a Hall mobility of 20 cm2/Vs. The third study was on the growth of Ti-doped MoS2 thin films by LPCVD. The successful doping was confirmed by Hall effect measurement and secondary ion mass spectrometry (SIMS). Different growth temperatures from 1000 to 700 ? were studied. Ti act as a donor in MoS2. The fourth study is on fluorine-doped SnO2 (FTO) which has many technological applications including solar cells and transistors. FTO was grown by an aqueous-spray-based method. The main objective was to compare the actual against the nominal concentration of fluorine using SIMS. The concentration of fluorine in the grown films is lower than the concentration of fluorine in the aqueous solution.?

Surveys conducted in this type of environment afford the opportunity to evaluate the practical use GPR under field conditions. Coastal environments are effective for this evaluation because they offer a host of conditions that GPR surveys do not normally encounter at one time. The relationship of the land to the coast, sub-surface conditions and reliable survey areas create a (")perfect storm(") to test how practical the use of GPR is in coastal environments. This research is a study of homestead cemeteries situated within the boundaries of Cape Canaveral Air Force Station (CCAFS), using GPR. The research has three main goals. The first is to utilize GPR to identify if there are any unknown burials at CCAFS. The second is to test the practical effectiveness of GPR in coastal environments where high water table, geology and saline conditions can limit the capability of the technique to resolve subsurface features. The third is to correlate data from the GPR survey with ethnographic information to enhance the protection and maintenance with what is already available for the cemeteries.Research methods include field-based geophysical data collection in addition to archival and ethnographic historic research. The field component, to which this research pertains, entailed an on-site GPR survey at the nine sites on Cape Canaveral Air Force Station. This was followed by analysis of the information from the survey using standard processing software. Subsequently, a thorough archival search was completed to link historic and ethnographic information with the archaeological data obtained on the cemeteries. The final result of this research was a report that provides a detailed description of the results of the GPR survey of the cemeteries at Cape Canaveral Air Force Station.

Though the human race has been fighting the malaria war for over 4,000 years and we have made great strides in eliminating malaria from many countries, we are treading on the edge of what could be another malaria epidemic primarily due to widespread drug resistance. There are documented cases of resistance for every known antimalarial in use today, including Artemisinins. It is critical that we open a new window of discovery in development of next generation antimalarials that circumvent current resistance paradigms. These compounds must attack new targets, have different speeds of action, and ideally possess powerful transmission blocking potential if they are to be successful antimalarial candidates. Screening endeavors historically focused on either synthetic or natural product libraries. Recent efforts have focused on combining privilege elements of natural products into synthetically tractable compounds to create hybrid libraries. To discover novel antimalarial pharmacophores, we have screened natural products derived from marine biodiversity as well as natural product-inspired synthetic libraries. Our phenotypic screening of 3,164 marine natural products from the Harbor Branch Oceanographic Institute, 56 high density combinatorial natural product based libraries from the Torrey Pines Institute for Molecular Studies, alkaloid, terpene, and macrocyclic libraries from Memorial Sloan Kettering Cancer Center, and 594 natural productinspired compounds from Asinex have identified several new selective antiplasmodial hit chemotypes. iv In this study, we have focused on compounds that exhibit cellular actions differing from current antimalarials. Two of the scaffolds, UCF 201 and 501, a spirocyclic chromane and a nitroquinoline, respectively, act early in the development cycle and block invasion. The alkaloid derived compound M03 blocks egress. UCF 501 cures malaria in the rodent model and significantly inhibits stage V gametocytogenesis. Given that discovery of transmission blocking agents are a priority in the malaria elimination strategies, this result is significant. This work is of high impact as it addresses a critical need in the field- next generation antimalarial scaffolds for malaria therapy and elimination campaign.

Rapidly acquiring drug resistance is threatening to exhaust our antimalarial drug arsenal and already requires the utilization of combination drug therapy in most cases. The global need for novel antimalarial chemical scaffolds has never been greater.Screening of natural product libraries is known to have higher hit rates than synthetic chemical libraries. This elevated hit rate is somewhat attributed to the greater biodiversity available in natural products. Marine life is the most biodiverse system on the planet, containing 34 of the 36 known phyla of life, and is expected to be a rich source of novel chemotypes. In collaboration with the Harbor Branch Oceanographic Institute in Ft. Pierce we have screened a library of over 2,800 marine macroorganism peak fractions against Plasmodium falciparum using the SYBR green I fluorescence-based assay. In this screening process we have identified six compounds from five novel chemical scaffolds all of which have low micromolar to submicromolar IC50 values and excellent selectivity indices. Additionally, one of these chemical scaffolds, the bis(indolyl)imidazole, was selected for further in vitro pharmacological and structure-activity relationship (SAR) profiling, key steps in the challenging process of identifying a new antimalarial drug lead compound.

About 10.9\% (960) of whom suffer from Hypoplastic Left Heart Syndrome (HLHS) - a subset of CHD where children are born with a single-ventricle (SV). A series of three surgeries are carried out to correct HLHS culminating in the Fontan procedure where venous flow returns passively to the lungs. The current configuration for the Fontan results in elevated Central Venous Pressure (CVP), inadequate ventricular preload, and elevated Pulmonary Vascular Resistance (PVR) leading to a barrage of disease. To alleviate these complications, a `self-powered' Fontan is suggested where an Injection Jet Shunt (IJS) emanating from the aorta is anastomosed to each pulmonary artery. The IJS attempts to reduce the central venous pressure, increase preload, and aid in pulmonary arterial growth by entraining the flow with a high energy source provided by the aorta. Previous computational studies on this concept with rigid vessel walls show mild success, but not enough to be clinically relevant. It is hypothesized that vessel wall deformation may play an important role in enhancing the jet effect to provide a larger exit area for the flow to diffuse while also being more physiologically accurate. A multiscale 0D-3D tightly coupled Computational Fluid Dynamics (CFD) with Fluid-Structure Interaction (FSI) model is developed to investigate the efficacy of the proposed `self-powered' Fontan modification. Several runs are made varying the PVR to investigate the sensitivity of IVC pressure on PVR. IVC pressure decreased by 2.41 mmHg while the rigid wall study decreased the IVC pressure by 2.88 mmHg. It is shown that IVC pressure is highly sensitive to changes in PVR and modifications to the Fontan procedure should target aiding pulmonary arterial growth as it is the main indicator of Fontan success.

The disruption of the steps required for the function of ribosomes represents a potential future target for pharmacological anti-cancer therapy. ABCE1 is an essential Fe-S protein involved in ribosomal function and is vital for protein synthesis and cell survival. Thus, ABCE1 is potentially a great therapeutic target for cancer treatment. Previously, cell biological, genetic, and structural studies uncovered the general importance of ABCE1, although the exact function of the Fe-S clusters was previously unclear, only a simple structural role was suggested. Additionally, due to the essential nature of ABCE1, its function in ribosome biogenesis, ribosome recycling, and the presence of Fe-S within ABCE1, the protein has been hypothesized to be a target for oxidative degradation by ROS and critically impact cellular function. In an effort to better understand the function of ABCE1 and its associated Fe-S cofactors, the goal of this research was to achieve a better biochemical understanding of the Fe-S clusters of ABCE1. The kinetics of the ATPase activity for the Pyrococcus abyssi ABCE1 (PabABCE1) was studied using both apo- (without reconstituted Fe-S clusters) and holo- (with full complement of Fe-S clusters reconstituted post-purification) forms, and is shown to be jointly regulated by the status of Fe-S clusters and Mg2+. Typically, ATPases require Mg2+, as is true for PabABCE1, but Mg2+ also acts as a unusual negative allosteric effector that modulates ATP affinity of PabABCE1. Comparative kinetic analysis of Mg2+ inhibition shows differences in the degree of allosteric regulation between the apo- and holo-PabABCE1 where the apparent Km for ATP of apo-PabABCE1 increases (>)30 fold from ~30 (&)#181;M to over 1 mM when in the presence of physiologically relevant concentrations of Mg2+. This effect would significantly convert the ATPase activity of PabABCE1 from being independent of cellular energy charge to being dependent on energy charge with cellular [Mg2+]. The effect of ROS on the Fe-S clusters within ABCE1 from Saccharomyces cerevisiae was studied by in vivo 55Fe labeling. A dose and time dependent depletion of ABCE1 bound 55Fe after exposure to H2O2 was discovered, suggesting the progressive degradation of Fe-S clusters under oxidative stress conditions. Furthermore, our experiments show growth recovery, upon removal of the H2O2, reaching a growth rate close to that of untreated cells after ~8 hrs. Additionally, a corresponding increase (~88% recovery) in the ABCE1 bound 55Fe (Fe-S) was demonstrated. Observations presented in this work demonstrate that the majority of growth inhibition, induced by oxidative stress, can be explained by a comparable decrease in ABCE1 bound 55Fe and likely loss of ABCE1 activity that is necessary for normal ribosomal activity. The regulatory roles of the Fe-S clusters with ABCE1 provide the cell a way to modulate the activity of ABCE1 and effectively regulate translation based on both cellular energy charge and the redox state of the cell. Intricate overlapping effects by both [Mg2+] and the status of Fe-S clusters regulate ABCE1's ATPase activity and suggest a regulatory mechanism, where under oxidative stress conditions, the translational activity of ABCE1 can be inhibited by oxidative degradation of the Fe-S clusters. These findings uncover the regulatory function of the Fe-S clusters with ABCE1, providing important clues needed for the development of pharmacological agents toward ABCE1 targeted anti-cancer therapy.

Many history textbooks contain the national story of Martin Luther King, Jr., Rosa Parks, the march to Selma, Alabama, and not much more. Classrooms across the United States follow this path of nationalizing the civil rights movement. This interpretation is only a small part of the civil rights crusade that existed throughout the United States, including in the state of Florida. Teaching only the national story, especially when the local exists, can ignore the human, ordinary element of this movement.The purpose of this phenomenological study was to describe the lived experience of central Florida teachers when teaching the civil rights movement. It is based on the theoretical assumptions that the national story is the only narrative being taught regarding the civil rights movement, and it sought to determine whether this is the case in the state of Florida, which incorporates the use of local history in its state standards. Data was collected through the use of surveys along with follow up, qualitative interviews. The sample size was 319 teachers of whom 65 responded to the survey, and eight personal interviews were conducted. Findings show that more than just Martin Luther King, Jr., and Rosa Parks are being taught, but it is still mostly the national story and not local, community history.Nine themes were identified, ranging from the impact of teachers, which builds upon previous research, to the negative opinion that teachers have for the texts being used, to the different content and timelines being used in social studies classrooms when teaching the civil rights movement. This data is important to educators, historians, administrators, and teachers because this is one of the first empirical studies on the subject of teaching the civil rights movement.

Findings revealed that increased access to books in these elementary schools did not have a statistically significant impact on reading motivation or reading achievement scores. In addition, teachers' demographic data, such as age, number of years teaching or highest degree held, did not impact their student reading achievement.

Tracking the various PoI allows for the composition of those points into 3D geometries which are unique to a given vehicle type. We demonstrate this technique using passive, simulated image based sensor measurements and three separate inertial track formulations. We demonstrate the capability to classify the 3D geometries in multiple transform domains (PCA (&) LDA) using Minimum Euclidean Distance, Maximum Likelihood and Artificial Neural Networks. Additionally, we demonstrate the ability to fuse separate classifiers from multiple domains via Bayesian Networks to achieve ensemble classification.
