Waste-to-energy is a sustainable method that may reduce bio-solids volume while also producing energy. In this research study bio-solids enters a bubbling bed gasifier and within the gasifier a synthesis gas is produced. This synthesis gas exits through the top of the gasifier and enters a thermal oxidizer for combustion. The thermal oxidizer has an innovative method of oxidizing the synthesis gas. The thermal oxidizer has two air injection sites and the possibility for aqueous ammonia injection for further NOx reduction. Most thermal oxidizers already include an oxidizer such as air in the fuel before it enters the thermal oxidizer; thus making this research and operation different from many other thermal oxidizers and waste-to-energy plants.The reduction in waste means less volume loads to a landfill. This process significantly reduces the amount of bio-solids to a landfill. The energy produced from the synthesis is beneficial for any municipality, as it may be used to run the waste-to-energy facility. The purpose of this study is to determine methods in which operators may configure future plants to reduce NOx emissions. NOx mixed with volatile organic compounds (VOC) and sunlight, produce ozone (O3) a deadly gas at high concentrations.This study developed a model to determine the best methods to reduce NOx emissions. Results indicate that a fuel-rich then fuel-lean injection scheme results in lower NOx emissions. This is because at fuel-rich conditions not all of the ammonia in the first air ring is converted to NOx, but rather a partial of the ammonia is converted to NOx and N2 and then the second air ring operates at fuel-lean which further oxidizes the remaining ammonia which converts to NOx, but also a fraction to N2. If NOx standards reach more stringency then aqueous ammonia injection is a recommended method for NOx reduction; this method is also known as selective non-catalytic reduction (SNCR).The findings in this study will allow operators to make better judgment in the way that they operate a two air injection scheme thermal oxidizer. The goal of the operator and the organization is to meet air quality standards and this study aims at finding ways to reduce emissions, specifically NOx.

Show moreFor a linear regression, the traditional technique deals with a case where the number of observations n more than the number of predictor variables p (n(>)p). In the case n(<)p, the classical method fails to estimate the coefficients. A solution of this problem in the case of correlated predictors is provided in this thesis. A new regularization and variable selection is proposed under the name of Sparse Ridge Fusion (SRF). In the case of highly correlated predictor , the simulated examples and a real data show that the SRF always outperforms the lasso, elastic net, and the S-Lasso, and the results show that the SRF selects more predictor variables than the sample size n while the maximum selected variables by lasso is n size.

Show moreI confronted empirical habitat data (1994-2004) and population data (1988-2005) with ecological theory on habitat dynamics, recruitment, survival, and dispersal to develop predictive relationships between landcover variation and population dynamics. I focus on Florida Scrub-Jays, although one chapter presents a model for the potential influence of habitat restoration on viability of the Florida Grasshopper Sparrow. Both species are unique to Florida landscapes that are dominated by shrubs and grasses and maintained by frequent fires. Both species are declining, even in protected areas, despite their protected status. I mapped habitat for both species using grid polygon cells to quantify population potential and habitat quality. A grid cell was the average territory size and the landcover unit in which habitat-specific recruitment and survival occurred. I measured habitat-specific recruitment and survival of Florida Scrub-Jays from 1988-2008. Data analyses included multistate analysis, which was developed for capture-recapture data but is useful for analyzing many ecological processes, such as habitat change. I relied on publications by other investigators for empirical Florida Grasshopper Sparrow data. The amount of potential habitat was greatly underestimated by landcover mapping not specific to Florida Scrub-Jays. Overlaying east central Florida with grid polygons was an efficient method to map potential habitat and monitor habitat quality directly related to recruitment, survival, and management needs. Most habitats for both species were degraded by anthropogenic reductions in fire frequency. Degradation occurred across large areas. Florida Scrub-Jay recruitment and survival were most influenced by shrub height states. Multistate modeling of shrub heights showed that state transitions were influenced by vegetation composition, edges, and habitat management. Measured population declines of 4% per year corroborated habitat-specific modeling predictions. Habitat quality improved over the study period but not enough to recover precariously small populations. The degree of landcover fragmentation influenced mean Florida Scrub-Jay dispersal distances but not the number of occupied territories between natal and breeding territories. There was little exchange between populations, which were usually further apart than mean dispersal distances. Florida Scrub-Jays bred or delayed breeding depending on age, sex, and breeding opportunities. I show an urgent need also for Florida Grasshopper Sparrow habitat restoration given that the endangered bird has declined to only two sizeable populations and there is a high likelihood for continued large decline. A major effect of habitat fragmentation identified in this dissertation that should apply to many organisms in disturbance prone systems is that fragmentation disrupts natural processes, reducing habitat quality across large areas. Humans have managed wildland fire for > 40,000 years, so it should be possible to manage habitat for many endangered species that make Florida's biodiversity unique. This dissertation provides methods to quantify landscape units into potential source and sink territories and provides a basis for applying adaptive management to reach population and conservation goals.

Macroscopic optical elements are easily fabricated using grinding and polishing techniques, but few methods exist for inexpensive fabrication of micro optical elements. In this work we present an innovative technique termed Additive Lithography that makes use of binary masks and controlled partial exposures to sculpt photoresist into the desired optical surface relief profile. We explore various masking schemes for fabricating a variety of optical elements with unprecedented flexibility and precision. These masking schemes used in conjunction with the additive lithographic method allows us to carefully control the photoresist exposure and reflow processes for fabricating complex aspheric lens elements, including aspheric elements whose fabrication often proves highly problematic. It will be demonstrated that employing additive lithography for volume sculpting followed by controlled reflow can also allow us to fabricate refractive beam shaping elements. Finally we will discuss the dry etching techniques used to transfer these optical elements into the glass substrate. Thus the additive lithography technique will be demonstrated as an inexpensive, high throughput and efficient process in the fabrication of micro optical elements.

This process of color change is a consequence of structural changes within the compound, such as cis-trans photo-isomerization. Some examples of photochromic compounds are spiropyrans, spirooxazines, diarylethenes and azobenzenes. These compounds have been extensively studied for decades, and are used in various applications such as biomedicine, chemical sensors and harvesting solar energy. However, majority of photochromic compounds are initially activated by ultraviolet (UV) light. The use of UV light is harmful for biological applications and photo-degrade the compound over repeated use. To overcome these limitations, a new class of reversible photochromic compound was introduced, called metastable-state photoacid (mPAH). In brief, mPAH is a photochromic compound which can photo-dissociate its protons under visible light and can thermally re-capture the released protons efficiently in the dark. Based on this unique property, in this research, we (1) synthesized different mPAH, and (2) studied and characterized their physicochemical (acidity, kinetics, and optical) properties. Additionally, we (3) applied different visible light activated mPAHs towards light controllable polymeric-based ion-selective optodes for detection of calcium ions and sodium ions, and modulate fluorescence with pH. The research presented herein opens new avenues towards the synthesis of mPAH derivatives and could be applied to any proton-transfer process related applications which requires wireless controllability with high sensitivity.

This dissertation reports on further innovations by presenting novel solid state laser systems based on the volume Bragg gratings (VBGs) and the newly developed holographic phase mask (HPMs) for brightness enhancement, dual wavelength operation, and mode conversion. First, a new optical element was created by pairing the HPM with two surface gratings creating an achromatic holographic phase mask. This new optical device successfully performed transverse mode conversion of multiple narrow line laser sources operating from 488 to 1550 nm and a broadband mode locked femtosecond source with no angular tuning. Also, two types of HPMs were tested on high power Yb fiber lasers to demonstrate high energy mode conversion.Secondly, the effects of implementing VBGs for brightness enhancement of passively Q-switched systems with large Fresnel numbers was investigated. Implementing VBGs for angular mode selection allowed for higher pulse energies to be extracted without sacrificing brightness and pulse duration. This technique could potentially be applied to construct compact cavities with 1 cm diameter beams and nearly diffraction limited beam quality.Lastly, a spectral beam combining approach was applied to create Tm3+ and Yb3+ based narrowband dual-wavelength pump sources for terahertz generation, using VBGs as frequency selectors and beam combiners. Comparison of pulse duration and synchronization was done between passive and active Q-switching operation. Show moreFor many years, successful applications of robotics were the domain of controlled environments, such as industrial assembly lines. Such environments are custom designed for the convenience of the robot and separated from human operators. In recent years, advances in artificial intelligence, in particular, deep learning and computer vision, allowed researchers to successfully demonstrate robots that operate in unstructured environments and directly interact with humans. One of the major applications of such robots is in assistive robotics. For instance, a wheelchair mounted robotic arm can help disabled users in the performance of activities of daily living (ADLs) such as feeding and personal grooming. Early systems relied entirely on the control of the human operator, something that is difficult to accomplish by a user with motor and/or cognitive disabilities. In this dissertation, we are describing research results that advance the field of assistive robotics. The overall goal is to improve the ability of the wheelchair / robotic arm assembly to help the user with the performance of the ADLs by requiring only high-level commands from the user. Let us consider an ADL involving the manipulation of an object in the user's home. This task can be naturally decomposed into two components: the movement of the wheelchair in such a way that the manipulator can conveniently grasp the object and the movement of the manipulator itself. This dissertation we provide an approach for addressing the challenge of finding the position appropriate for the required manipulation. We introduce the ease-of-reach score (ERS), a metric that quantifies the preferences for the positioning of the base while taking into consideration the shape and position of obstacles and clutter in the environment. As the brute force computation of ERS is computationally expensive, we propose a machine learning approach to estimate the ERS based on features and characteristics of the obstacles. This dissertation addresses the second component as well, the ability of the robotic arm to manipulate objects. Recent work in end-to-end learning of robotic manipulation had demonstrated that a deep learning-based controller of vision-enabled robotic arms can be thought to manipulate objects from a moderate number of demonstrations. However, the current state of the art systems are limited in robustness to physical and visual disturbances and do not generalize well to new objects. We describe new techniques based on task-focused attention that show significant improvement in the robustness of manipulation and performance in clutter.

Show moreLaser additive manufacturing (LAM) is an emerging technology capable of fabricating complex geometries not possibly made by investment casting methods for gas turbine applications. LAM techniques consist of building parts in a layer-by-layer process by selectively melting metal powders. In the present study, a mock leading edge segment of a turbine blade fabricated by LAM of Inconel 718 powders is investigated. For this particular design, the traditional showerhead film cooling holes have been replaced by two strips containing engineered-porous regions with the purpose of simulating the effect of transpiration cooling. Transpiration cooling has been considered a promising external convective cooling method capable of providing a more uniform film and higher adiabatic film cooling effectiveness than conventional discrete film cooling. In addition, many studies have shown that this technique can yield high firing temperatures with much less coolant consumption than discrete film cooling. In this current study, adiabatic film cooling effectiveness is investigated by means of mass transfer using pressure sensitive paint (PSP). The experiments are conducted for blowing ratios ranging between M = 0.03 and M = 0.28 for a nominal density ratio of 1.5. The density ratio is obtained by using air as the mainstream flow and CO2 as the secondary flow (or coolant source). Results indicate higher coverage and film cooling effectiveness when increasing blowing ratio at the expense of higher pressure drop. In addition, the experimental results are compared to numerical analyses performed using steady state Reynolds Average Navier Stokes (RANS) simulations.

Show moreArtificial beach nourishment, the most common method to mitigate coastal erosion in the United States, is also considered the most ecologically friendly alternative for shoreline stabilization. However, this habitat alteration has the potential to impact nesting marine turtles and developing hatchlings. The first objective of this study was to determine how nourishing beaches with two different design templates affects loggerhead (Caretta caretta) and green turtle (Chelonia mydas) nesting success, the ratio of nests to the total number of nests and non-nesting emergences, and reproductive success, the ratio of hatched and emerged hatchlings to the total number of eggs deposited. Two types of restoration designs exist along the southern Brevard County, FL coastline, which supports some of the highest density loggerhead and green turtle nesting worldwide. Since 2005, approximately 35 kilometers of beach have undergone 1) full-scale restoration (typically called nourishment), where sand was added above and below the mean high tide line (2005, 2010) or 2) dune restoration, where sand was placed on the dune (2005, 2006, 2008, 2009). To quantify the effects of these restoration types, we used a Before-After-Control-Impact-Paired Series (BACIPS) model, which tests for significance between the difference in nesting success rates at the impact (engineered) and control sites (natural beach) before and after restoration (?). For loggerheads, there was a significant difference in ? after dune restoration during the years of construction (2005, 2006, 2008, and 2009; p(<)0.001) and one year post-construction (2007; p(<)0.05 and 2010; p(<)0.001). After full-scale restoration, there was a significant difference in ? during the years of construction (2005 and 2010; p (<)0.001) and one year post-construction (2006; p(<)0.001). For green turtles, there was a significant difference in ? after dune restoration during two of the four years of construction (2006; p(<)0.05 and 2008; p(<)0.01) and one of the two one-year post-construction years (2010; p(<)0.05). After full-scale restoration, the significant difference in ? lasted every season (2005-2010; p(<)0.001). There were no significant differences in ? for loggerhead or green turtle reproductive success rates after either type of restoration. The second objective was to use the different restoration designs to study what beach characteristics function as loggerhead nesting cues to explain why altering the beach decreases nesting success rates. We examined beach elevation and slope, sand moisture content, sand grain size, beach width, and distance traveled. Logistic regression model selection found all variables were important (R2=0.75). Further examination of trends, with each crawl divided into quarters, found beach slope served as a nesting cue. In all study sites except one, when turtles false crawled, the beach flattened out in the final quarter of the crawl. Conversely, in nesting emergences, the final quarter rose at a steeper slope than the previous quarter. Additionally, model selection found variables important in nest site selection were also important in hatching (R2=0.44) and emergence (R2=0.45) success. These results offer new insight into how and why marine turtle nesting patterns change after artificial nourishment, providing information necessary to nourish beaches in a more (")turtle-friendly(") manner. ?

Show moreThe aim of this dissertation is to gain a better understanding of the unique electronic structure of lead salt quantum dots (QDs) and its influences on the nonlinear optical (NLO) properties as well as the time dynamics of the photogenerated charge carriers. A variety of optical techniques such as Z-scan, two-photon excited fluorescence and time-resolved pump probe spectroscopy are used to measure these properties. The one-photon as well as the degenerate and nondegenerate two-photon absorption (2PA) spectra are measured and the electronic wave functions from a four-band envelope function formalism are used to model the results. We observe local maxima in the 2PA spectra for QD samples of many different sizes at energies where only 1PA is predicted by the model. This is similar to the previously measured transitions in the 1PA spectra which are not predicted by the model but accrue at the energies of the two-photon allowed transitions. Most importantly we observe 2PA peaks for all samples at the energy of the first one-photon allowed transition. This result can only be understood in terms of symmetry breaking and therefore is strong evidence that other transitions, not predicted by the model if the selection rules are left intact, also have the origin in the lifted spatial symmetry of the wave functions. On the other hand, the uniquely symmetric eigenenergies of these quantum-confined energy states in the conduction and valance bands explain the observed trend toward larger two-photon cross-sections as the quantum confinement is increased in smaller QDs. Moreover, this unique feature is shown to reduce the possible relaxation channels for photoexcited carriers, which is confirmed experimentally by the reduced carrier relaxation rate as compared to CdSe QDs which lack this symmetry. Carrier multiplication (CM), a process in which several electrons are excited by the absorption of a single photon is studied in PbS QDs. We show that for PbS QDs with radius smaller than 2.5 nm the parameters of CM get very close to the theoretical optimum. Next-generation solar cells operating under these ideal conditions could potentially have conversion efficiency of up to 42%. This compares favorably to the 30% efficiency limit of a single junction silicon solar cell.

Amphibians provide excellent model organisms for investigating the interplay between these evolutionary forces because amphibians are generally characterized by limited dispersal abilities, high philopatry, and are obligately associated with the areas around suitable habitats (e.g. breeding ponds). Thus, on relatively small geographic scales, the relative effects of all of these evolutionary forces can be studied together. Here, we study the interaction of migration, genetic drift, natural selection, and historical process in the ornate chorus frog (Pseudacris ornata). We report the development and characterization of 10 polymorphic microsatellite genetic markers. Number of alleles per locus ranged from 2 to 21 averaging 9.2 and expected heterozygosities ranged from 0.10 to 0.97 averaging 0.52. However, in an analysis of two populations, three locus-by-population comparisons exhibited significant heterozygote deficiencies and indicated that null alleles may be present some loci. Furthermore, we characterized genetic structure and historical biogeographic patterns in P. ornata using these microsatellite markers along with mitochondrial DNA sequence data. Our data indicate that in these frogs, migration may play a large role in determining population structure as pairwise estimates of FST were relatively small ranging from 0.04 to 0.12 (global FST = 0.083). Additionally, we observed an overall pattern of isolation-by-distance in neutral genetic markers across the species range. Moreover, our data suggest that the Apalachicola River basin does not impede gene flow in P. ornata as it does in many vertebrate taxa. Interestingly, we identified significant genetic structure between populations separated by only 6 km. However, this fine scale genetic structure was only present in the more urbanized of two widespread sampling localities. Finally, in this study, we demonstrated that there was a significant correlation between the frequency of green frogs and latitude. There was a higher frequency of green frogs in southern samples and a lower frequency of green frogs in northern samples. However, when we interpreted this phenotypic cline in light of the overall pattern of isolation-by-distance, it was apparent that the neutral evolutionary forces of genetic drift and migration could explain the cline, and the invocation of natural selection was not necessary.

We have only recently begun to address how manatees use some of these resources based on physiological or reproductive strategies. In this study, I quantified patterns of habitat use during seasonal and diel periods for different sex and reproductive manatee classes using data from a radio-telemetry study conducted by the Florida Fish and Wildlife Conservation Commission during 1991-1996. I used five environmental geographic data layers: bathymetry, distance to seagrass, distance to shoreline, distance to warm water refuge sites, and distance to fresh water sources, to discriminate seasonal and diel habitat use patterns for different manatee classes: males (M), females with calves (FWC), and females without calves (FNC). Mean occupancy values were calculated for environmental variable locations and seasonal, diel, and manatee class differences were tested using a Multi-Response Permutation Procedure (MRPP). Nonmetric Multidimensional Scaling (NMS) was used to visualize the ordination patterns of the manatee classes and to assess importance of correlated environmental variables. Significant differences in habitat use were noted between summer and winter based on distances to warm water, seagrass, and fresh water sources but similar habitat use patterns were exhibited within summer diel periods among manatee classes. All manatee classes appeared to have used a higher proportion of locations closer in proximity to seagrass at night than day in winter indicating a disproportionate difference in feeding bouts between diel periods. These differences may be attributed to adjusting feeding strategies to reduce thermoregulatory costs or to decrease human interactions. Differences in patterns were exhibited for the winter diel periods specifically for the FWC manatee classes during winter days. FWC had a higher proportion of locations within the warm water refuges during the day indicating a possible trade off situation between food consumption and thermal exposure. This study demonstrates coarse and fine scale patterns of variation in habitat use for manatees both seasonally and daily within winter. It also suggests that during winter months, manatees were not just utilizing their habitat but they appeared to have preferences and selection for certain habitat types. Recovery of a species is greatly enhanced when patterns of habitat use within the species' environment has been clearly defined. Understanding more specifically what types of habitats manatees choose might allow management to adjust strategies for protection of key habitats while encouraging further recovery of this species.

Show moreIt is desirable to modify the spectral signature of a surface, particularly in the infrared (IR) region of the electromagnetic spectrum. To alter the surface signature in the IR, two methods are investigated: thin film application and antenna array application. The former approach is a common and straightforward incorporation of optically-thin film coatings on the surface designated for signature modification. The latter technique requires the complex design of a periodic array of passive microantenna elements to cover the surface in order to modify its signature. This technology is known as frequency selective surface (FSS) technology and is established in the millimeter-wave spectral regime, but is a challenging technology to scale for IR application. Incorporation of thin films and FSS antenna elements on a surface permits the signature of a surface to be changed in a deterministic manner. In the seminal application of this work, both technologies are integrated to comprise a circuit-analog absorbing IR FSS. The design and modeling of surface treatments are accomplished using commercially-available electromagnetic simulation software. Fabrication of microstructured antenna arrays is accomplished via microlithographic technology, particularly using an industrial direct-write electron-beam lithography system. Comprehensive measurement methods are utilized to study the patterned surfaces, including infrared spectral radiometry and Fourier-transform infrared spectrometry. These systems allow for direct and complementary spectral signature measurements--the radiometer measures the absorption or emission of the surface, and the spectrometer measures its transmission and reflection. For the circuit-analog absorbing square-loop IR FSS, the spectral modulation in emission is measured to be greater than 85% at resonance. Other desirable modifications of surface signature are also explored; these include the ability to filter radiation based on its polarization orientation and the ability to dynamically tune the surface signature. An array of spiral FSS elements allows for circular polarization conditioning. Three techniques for tuning the IR FSS signature via voltage application are explored, including the incorporation of a pn junction substrate, a piezoelectric substrate and a liquid crystal superstrate. These studies will ignite future explorations of IR FSS technology, enabling various unique applications.

This study was intended to provide information for higher education practitioners, faculty, and administrators to help them better understand the expectations and current trends of National Merit Finalists. The problem was to determine how to increase recruitment and retention while decreasing the attrition rates of these highly desirable students. The importance of this study includes identifying trends that may aid in future recruitment efforts for National Merit Finalists; finding the causes of dissatisfaction towards the University among these students; and identifying specific areas in which to alleviate those dissatisfactions. The results will hopefully provide insight into specific recruitment, services, and programming options for these students. The study examined data that was collected from the University of Central Florida's Burnett Honors College database known as FileMaker 8.0. The data examined characteristics such as grade point averages (high school and college); valedictorian and salutatorian status; test scores (SAT and ACT); Honors in the Major (undergraduate thesis) students; Honors and university status (withdrawn, probation, removed, disqualified, enrolled, graduated); Honors college attrition; university attrition; ethnicity; gender ratios; majors; and, prestigious scholarships awarded in college (such as the Rhodes, Truman, Marshall). The actual size of the sample was one hundred ninety-eight National Merit Finalists. Data was also collected from a survey given to all University of Central Florida National Merit Finalists. Descriptive statistics were reported for each of the components examined. This data examined the types of scholarship packages that National Merit Finalists were offered; the reasons students chose the University of Central Florida over other universities; the college recruitment process; hours studied for the PSAT; siblings; perceptions on being a National Merit Finalist; the number of times students changed their majors; job status; transportation; computer attainment; disabilities; and the potential disadvantages of being labeled as a National Merit Finalist. The data could be utilized to examine the trends of our National Merit Finalists, in order to see what is working and what is not in terms or recruitment and retention; and also to further examine what these students want from their institutions. Findings indicated that problems exist in regard to the following: the recruitment of female and minority National Merit Finalists; males historically score higher on the SAT than females; decreasing the attrition rates of this population at the University of Central Florida; the majority of National Merit Finalists at the University of Central Florida come from Florida; the majority of National Merit Finalists at the University of Central Florida do not tend to be high school salutatorians or valedictorians; high school counselors seem to be the least effective tool for recruiting National Merit Finalists at the University of Central Florida; and the majority of National Merit Finalists at the University of Central Florida did not study at all for the PSAT test. However, the University of Central Florida is extremely competitive with other institutions of higher education with regard to scholarship packages. Results also revealed the following: the SAT is a more widely accepted tool for determining NMSC status as opposed to the ACT; the majority of National Merit Finalists have a GPA between 3.600 and 3.999 at the University of Central Florida; the University of Central Florida is succeeding in making its National Merit Finalists feel special during the recruitment process; the most influential reason that National Merit Finalists are choosing UCF is based upon the financial scholarship packages they are offered; and the majority of National Merit Finalists at the University of Central Florida do not feel that there are disadvantages toward being labeled as such. This data provides a basis for further research on National Merit Finalists trends, attrition, and retention. Practical considerations are revealed in the data that will influence future recruitment methods and lead to higher retention rates and increased student satisfaction. Several other recommendations are made to conduct further research studies on the trends, attrition, and retention rates of National Merit Finalists.

Compounding the problem is the emergence of drug-resistant strains due to overuse of outdated treatments. New compounds with antiplasmodial activity are needed to be developed as effective drugs against malaria. The hypothesis for this project is that marine microorganisms have a high likelihood of yielding novel antiplasmodial chemotypes because of their high diversity, which has not yet been explored for antimalarial development. In this project, microbes harvested and fermented by the Harbor Branch Oceanographic Institute in Fort Pierce, Florida were explored as sources for antiplasmodial natural products. Using a SYBR Green I fluorescence-based assay, 1,000 microbial extracts were screened for inhibition of the multidrug-resistant Plasmodium falciparum strain Dd2. Dose-response analysis was performed on 46 fractions from isolates whose extracts demonstrated [greater-than or equal to] 70% inhibition of Dd2 at 1 [micro]g/mL. To evaluate cytotoxicity, the MTS cell viability assay was used to calculate IC50 of extracts from active isolates in NIH/3T3 embryonic mouse fibroblasts. Several extracts demonstrated low IC50 in Dd2 and high IC50 in 3T3, suggesting that they contain potential lead antimalarial compounds. Extracts with high selectivity indices (potent plasmodial inhibition with low mammalian toxicity) have been prioritized for dereplication, with the goal of identifying novel active components that can be developed as antimalarial drugs.

We present a data mining framework to detect malicious programs. We collected, analyzed and processed several thousand malicious and clean programs to find out the best features and build models that can classify a given program into a malware or a clean class. Our research is closely related to information retrieval and classification techniques and borrows a number of ideas from the field. We used a vector space model to represent the programs in our collection. Our data mining framework includes two separate and distinct classes of experiments. The first are the supervised learning experiments that used a dataset, consisting of several thousand malicious and clean program samples to train, validate and test, an array of classifiers. In the second class of experiments, we proposed using sequential association analysis for feature selection and automatic signature extraction. With our experiments, we were able to achieve as high as 98.4% detection rate and as low as 1.9% false positive rate on novel malwares.

Thus, this material is a good candidate for processing development in the direct metal laser sintering (DMLS) branch of additive manufacturing. The chemistry and microstructure of this alloy processed via DMLS was compared to its conventionally cast counterpart through various heat treatments as part of a characterization effort. The investigation utilized optical microscopy, scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-Ray diffractometry (XRD), energy dispersive X-Ray spectroscopy (EDS) and glow discharge atomic emission spectrometry (GDS) techniques. DMLS processed samples contained a layered microstructure in which the prior austenite grain sizes were relatively smaller than the cast and annealed prior austenite grain size. The largest of the quantifiable DMLS prior austenite grains had an ASTM grain size of approximately 11.5-12 (6.7?m to 5.6?m, respectively) and the cast and annealed prior austenite grain size was approximately 7-7.5 (31.8?m to 26.7?m, respectively), giving insight to the elevated mechanical properties of the DMLS processed alloy. During investigation, significant amounts of retained austenite phase were found in the DMLS processed samples and quantified by XRD analysis. Causes of this phase included high nitrogen content, absorbed during nitrogen gas atomization of the DMLS metal powder and from the DMLS build chamber nitrogen atmosphere. Nitrogen content was quantified by GDS for three samples. DMLS powder produced by nitrogen gas atomization had a nitrogen content of 0.11 wt%. A DMLS processed sample contained 0.08 wt% nitrogen, and a conventionally cast and annealed sample contained only 0.019 wt% nitrogen. In iron based alloys, nitrogen is a significant austenite promoter and reduced the martensite start and finish temperatures, rendering the standard heat treatments for the alloy ineffective in producing full transformation to martensite. Process improvements are proposed along with suggested future research.

Mojave Rattlesnakes, Crotalus scutulatus, are known to have high intraspecific venom variation, but the geographic extent of the variation and factors influencing venom evolution are poorly understood. Three primary venom types have been described in this species based on the presence (Type A) or absence (Type B) of a neurotoxic phospholipase A2 called Mojave toxin and an inverse relationship with the presence of snake venom metalloproteinases (SVMPs). Individuals that contain both Mojave toxin and SVMPs, although rare, are the third, and designated Type A + B. I sought to describe the proteomic and transcriptomic venom diversity of C. scutulatus across its range and test whether diversity was correlated with genetic or environmental differences. This study includes the highest geographic sampling of Mojave Rattlesnakes and includes the most venom-gland transcriptomes known for one species. Of the four mitochondrial lineages known, only one was monophyletic for venom type. Environmental variables poorly correlated with the phenotypes. Variability in toxin and toxin family composition of venom transcriptomes was largely due to differences in transcript expression. Four of 19 toxin families identified in C. scutulatus account for the majority of differences in toxin number and expression variation. I was able to determine that the toxins primarily responsible for venom types are inherited in a Mendelian fashion and that toxin expression is additive when comparing heterozygotes and homozygotes. Using the genetics to define venom type is more informative and the Type A + B phenotype is not unique, but rather heterozygous for the PLA2 and/or SVMP alleles. Intraspecific venom variation in C. scutulatus highlights the need for fine scale ecological and natural history information to understand how phenotypic diversity is generated and maintained geographically through time.

However, because scrub burns infrequently such early-successional, high-quality habitat is extremely limited and competition for it as breeding space is likely intense. Because some birds live long enough to experience habitat succession, FLSJs also occupy later-successional overgrown scrub, even though both survival and fecundity decline. Although immigration rates into later-successional habitat decline, some birds settle there, perhaps to avoid competition. Prior to dispersal into new breeding territories, most non-breeders engage in pre-dispersal forays, which occur before and immediately after the breeding season. Because FLSJ territories occur across a gradient of post-fire succession, and young birds make frequent forays beyond their natal territories, and are highly visible while doing so, they are ideal models to test how individual and environmental factors drive habitat preference when exploring a post-fire mosaic. I investigated how individual behavioral phenotype, natal territory condition, and body condition relative to conspecifics influence extra-territorial foray behavior across a habitat mosaic that includes various time-since-fire (TSF) intervals. My study system was a population of individually marked FLSJs on 1,214-ha of managed scrub at Archbold Biological Station (Highlands County, Florida). I measured off-territory movements of 114 yearling birds in three annual cohorts and quantified habitat preference using fine-scale fire history records. These data, paired with behavioral assays and morphometric records for each individual FLSJ, allowed me to create and compare models of exploratory behavior underlying searching patterns during pre-breeding forays. My results indicate significant variation in habitat preference by individual FLSJs during forays, driven by an individual's behavioral phenotype and the TSF of its natal territory.

Show moreIn this thesis the notion of a theoretical approach to the beginning stages of designing lighting for a theatrical production will be discussed. The topic being researched is: How a theoretical approach to entering the design process will enhance the final lighting design. The target audience for this study is theatrical lighting designers. A theoretical approach, in this case to the beginning of the design process, could be described as utilizing current dramatic theories to develop a better understanding for the design of this production. In order to better understand this topic one would need to know how the process of lighting design is typically created and where the theoretical approach is implemented. An issue with this approach is that the short period allowed for the design process does not allow sufficient time to utilize a theoretical approach in a real world setting. A way of determining if this process is effective is through personal self review. Journaling and discussion with my advisor for this production will be the method of data collection. The method of validation will be a self reflection at the end of the final performance. An issue with the collection process is its reliance on personal opinions, including the author's. There are no ethical issues relating to this study. When applied, a theoretical approach to the design process will enhance the quality of the final lighting design through allowing the designer to be better prepared for a specific scene that he/she is struggling with.
