Show moreThis thesis investigates the optimal ex-ante mix of self-protection and self-insurance technologies employed to reduce the risk of biological invaders in the presence of exogenous variables within the probability and damage functions. This is accomplished by using a theoretical endogenous risk model that extends previously developed frameworks. This thesis contributes to the previous work in two ways. (1) employing a general framework with simultaneous decision making over self-protection and self-insurance, this thesis analyzes how each parameter including income, the costs of each activity, an exogenous factor that affects only the probability of an invasion, and a separate exogenous factor that affects only the damages influence the level of self-protection and self-insurance. (2) the comparative static results are derived in the benchmark case of risk neutral preferences and qualitatively compared to an extended case of risk averse preferences. The results of the analysis indicate that under risk neutral preferences the signs of the comparative statics are unambiguous while under risk aversion the results are not clearly defined. Thus, the ambiguity of signs in the latter case can be attributed to the unobservable utility terms present under risk averse preferences. Therefore, it can be concluded that the model is not capable of yielding a decision criteria that will hold universally because the results are dependent on the nature of the risk averse curve

Show moreMangrove ecosystems are critical to Florida, providing economic resources to humans, and untold ecological resources to estuarine organisms. In Florida's estuaries, mangrove ecosystems have suffered significant losses due to natural and human disturbances; these disturbances potentially leave mangrove communities vulnerable to invasion by the opportunistic exotic, Schinus terebinthifolius (Brazilian pepper). Prior experiments have suggested that Schinus terebinthifolius is unable to survive under marine conditions and poses no long term threat to mangrove systems. However, this contradicts field observations where Schinus terebinthifolius was found growing in the intertidal zone of Mosquito Lagoon alongside three native species of mangroves, Rhizophora mangle, Avicennia germinans and Laguncularia racemosa. The purpose of my study was to evaluate competition between the invasive S. terebinthifolius and these three native species of mangroves in the coastal estuarine system along the east coast of Central Florida in Canaveral National Seashore. The effects of competition were evaluated by testing the ability of S. terebinthifolius to: 1) chemically inhibit growth of mangrove propagules, 2) invade new coastal habitats by dispersing seeds in the water, 3) alter species richness and abundance of the flora when present in a mangrove system, and 4) recruit and survive in mangrove habitat. By better understanding the invasibility and impact of Schinus terebinthifolius on mangroves, coastal resource managers will be able to develop the most effective management strategies to prevent this exotic from altering the structure and productivity of the mangrove ecosystem.

Show moreInvasions of exotic species are one of the primary causes of biodiversity loss on our planet (National Research Council 1995). In the marine environment, all habitat types including estuaries, coral reefs, mud flats, and rocky intertidal shorelines have been impacted (e.g. Bertness et al. 2001). Recently, the topic of invasive species has caught the public's attention. In particular, there is worldwide concern about the aquarium strain of the green alga Caulerpa taxifolia (Vahl) C. Agardh that was introduced to the Mediterranean Sea in 1984 from the Monaco Oceanographic Museum. Since that time, it has flourished in thousands of hectares of near-shore waters. More recently, C. taxifolia has invaded southern Californian and Australian waters. Since the waters of Florida are similar to the waters of the Mediterranean Sea and other invasive sites my study will focus on determining potential invasion locations in Florida. I will look at the present distribution of C. taxifolia - native strain in Florida as well as the distribution of the whole genus around the state. During this study, I address three questions: 1) What is the current distribution of Caulerpa spp. in Florida? 2) Can I predict the location of potential Caulerpa spp. invasions using a set of environmental parameters and correlate them to the occurrence of the algae with the support of Geographic Information System (GIS) maps? 3) Using the results of part two, is there an ecological preferred environment for one or all Caulerpa spp. in Florida? To answer these questions, I surveyed 24 areas in each of 6 zones chosen in a stratified manner along the Floridian coastline to evaluate the association of potential indicators Caulerpa. Latitude, presence or absence of seagrass beds, human population density, and proximity to marinas were chosen as the 4 parameters expected to correlate to Caulerpa occurrences. A logistic regression model assessing the association of Caulerpa occurrence with measured variables has been developed to predict current and future probabilities of Caulerpa spp. presence throughout the state. Fourteen different species of Caulerpa spp. were found in 26 of the 132 sites visited. There was a positive correlation between Caulerpa spp. and seagrass beds presence and proximity to marinas. There was a negative correlation with latitude and human population density. C. taxifolia aquarium strain wasn't found. Percent correct for our model was of 61.5% for presence and 98.1% for absence. This prediction model will allow us to focus on particular areas for future surveys.

I experimentally tested dispersal behavior of individual mosquitofish under a range of laboratory conditions relevant to field situations. Preliminary experiments showed that gender, lighting conditions, hunger and acclimation time did not significantly affect net dispersal rate. Power analysis based on this preliminary experiment determined that 6 replicate fish were sufficient for each subsequent experiment; I used 24 fish, and each fish was tested one time. Three factors that potentially could affect net swimming rate were tested: habitat of origin (permanent vs. temporary waters), water depth (3-24 mm), and the interaction between water depth and leaf litter type (upland and wetland). Fish from a temporary pond dispersed significantly faster than fish from a permanent pond, and fish dispersed significantly faster in deeper water than in shallower water. However, leaf litter significantly inhibited fish dispersal at all depths tested. Based on these experiments, G. holbrooki disperse more readily through relatively open and deeper (several centimeters) pathways between habitats such as roadside ditches, drainage canals and trails in flooded conditions. My results are useful for understanding mosquitofish dispersal behavior based on the abiotic and biotic factors examined in this experiment. I predict that mosquitofish can spread from a point of introduction at about 800 m per day, given and unobstructed path of only > 6 mm depth.

Cuban Treefrogs thrive in disturbed areas and around buildings, and often breed in manufactured wetlands such as retention ponds and borrow pits. This study identified limnological, landscape, and biotic characteristics that discouraged pond use by Cuban Treefrogs and promoted use by native amphibian species. I sampled natural and manufactured ponds in Orange County, Florida, for one year, using standard methods to estimate the species richness and relative abundance of amphibians and their potential fish and macroinvertebrate predators. I determined the relationship between the presence of Cuban Treefrogs and twelve limnological (% vegetation, slope, pond age, pH, % dissolved oxygen, air temperature, water temperature, turbidity, conductivity, depth, perimeter, and area) and seven landscape characteristics (% canopy closure over ponds, building density, distance to nearest building, road density, distance to nearest road, distance to nearest forest stand, and % forest cover), plus five biotic factors (native amphibian richness and abundance, fish richness and abundance, and macroinvertebrate abundance). No relationship existed between native amphibian abundance or species richness and the presence or absence of Cuban Treefrogs. Ponds with a greater percentage of vegetation, large perimeters, and low pH and turbidity had greater native amphibian species richness. Cuban Treefrogs were more likely to be found in ponds with a greater percentage of aquatic vegetation and small perimeters. My results show that building large retention ponds containing low-turbidity water will restrict colonization by Cuban Treefrogs and maintain species richness of native amphibians.

We evaluated prostasin expression in 12 BPH specimens by immunohistochemistry, and evaluated the impact of prostasin silencing by siRNA on the expression of the inducible nitric oxide synthase (iNOS), intercellular adhesion molecule-1 (ICAM-1), and cyclin D1, as well as on cell proliferation and invasion, using the BPH-1 human prostate epithelial cell line model. Prostasin expression was localized in the glands of BPH tissues by immunohistochemistry, in either the tall columnar-shaped or the flattened epithelial cells. We silenced prostasin expression by >50% at both the mRNA and protein levels using siRNA in the BPH-1 human prostate epithelial cell line, and this silencing of prostasin expression was associated with an induction of iNOS and ICAM-1 expression and a down-regulation of cyclin D1 expression. The protein expression of EGFR, a putative prostasin substrate, was not affected by prostasin silencing in this cell line. The prostasin-silenced cells displayed a reduced cell proliferation rate and reduced invasiveness, cell behaviors regulated by cyclin D1, iNOS, and ICAM-1 in the BPH-1 cells. We believe that this down-regulation of cyclin D1 is due to prostasin's augmentative effect on iNOS. We also believe that the decrease in cell motility is due to an increase in iNOS and ICAM-1 as well as a decrease in cyclin D1, since all of these molecules can play a role in cell motility. In conclusion, Prostasin is somehow involved in the regulation of inflammatory gene expression (iNOS and ICAM-1) in prostate epithelial cells, as well as cyclin D1 expression, cell proliferation and invasion, involving molecular mechanisms different than those in the prostate cancer cells. These studies suggest that prostasin is a player in the glandular components of benign prostatic hyperplasia.

This snail is a serious pest of rice crops in Asia, but little is known about its interactions within Florida ecosystems. Possible competitive exclusion of the native congener (P. paludosa) is a concern because it is the almost exclusive prey of the federally endangered Everglades snail kite (Rostrhamus sociabilis). My research consisted of three parts. First, to identify possible methods to control P. canaliculata in the egg stage, I experimentally evaluated the effects of photoperiod, substrate availability, and food type and availability on hatching success and hatchling survival. None of these factors significantly affected hatching or survival, suggesting that P. canaliculata could invade many water bodies. Second, I used a randomized block design to assess the separate and combined effects of population density and food availability on growth and spawning of P. canaliculata. Food availability but not density significantly affected growth; snails fed less food grew less, but neither factor affected spawning. Third, I used a replacement series design to assess competition between P. canaliculata and P. paludosa under food limitation. For both species, snails fed less grew significantly less, and snails exposed to greater congener densities spawned less than when exposed to conspecifics only. While my results predict that P. paludosa should exclude P. canaliculata, previous literature and current invasions suggest that the exotic snail may co-exist with its North American congener.

However many examinations of invasion success ignore the contribution of dispersal to patterns of invasion and focus only on the local environmental/habitat factors. This study examines the interaction of geospatial characteristics, that may influence dispersal, and local environmental factors, that may govern successful occupation, on the likelihood of invasion (invasability) of wetlands within an agriculturally modified landscape. I examined the current invasion of seasonal wetlands in south-central Florida ranchland by non-native apple snails (Pomacea maculata (Ampullariidae)) as a model system for understanding this interaction. I surveyed spatial occurrence of P. maculata in 171 wetlands in 2014 and found they occurred in 43% of wetlands surveyed. I evaluated how occurrence was related to geospatial variables (proximity to propagule sources, shoreline complexity, interwetland distance, elevation, area and ditch presence) and wetland characteristics (pH, water hardness, conductivity and soil type) for 95 wetlands. Presence of ditch connections and more neutral water pH were associated with P. maculata occurrence. I did not find evidence that Euclidean distance and minimum ditch distance were associated with P. maculata occurrence. I also performed a 5 month field experiment where I translocated snails to previously occupied and non-occupied wetlands and measured snail survival and growth (20 wetlands from November to March). This experiment evaluated if non-occurrence during survey was more likely to be associated with unfavorable habitat conditions or dispersal limitation. Wetland pH and water hardness explained variation in P. maculata survival, and wetland pH best explained growth. I did not find evidence that prior occupancy affected the snail survival and growth, which suggests previously unoccupied wetlands are due to dispersal limitation. These results emphasize that man-made conduits can increase permeability of the landscape, facilitating the dispersal and introduction of nonnative species and the need for the inclusion of dispersal metrics in understanding invasive species distribution.

The slightest unexpected movements could offset the readings of the automatic Blood Pressure meter by a large amount or render the readings totally meaningless. Every effort must be taken to avoid subjecting the body of the patient or the patient's surroundings to motion for obtaining a reliable reading. But there are situations in which we need Blood Pressure Measurements with the patient or his surroundings in motion; for instance in an ambulance while a patient is being transported to a hospital. In this thesis, we present a technique to reduce the effect of motion artifact from Blood Pressure measurements. We digitize the blood pressure waveform and use Digital Signal Processing Techniques to process the corrupted waveform. We use the differences in frequency spectra of the Blood Pressure signal and motion artifact noise to remove the motion artifact noise. The motion artifact noise spectrum is not very well defined, since it may consist of many different frequency components depending on the kind of motion. The Blood Pressure signal is more or less a periodic signal. That translates to periodicity in the frequency domain. Hence, we designed a digital filter that could take advantage of the periodic nature of the Blood Pressure Signal waveform. The filter is shaped like a comb with periodic peaks around the signal frequency components. Further processing of the filtered signal: baseline restoration and level shifting help us to further reduce the noise corruption.

Show morePopulations of Crassostrea virginica within Mosquito Lagoon, Florida have recently undergone significant die-offs, which are a subject of major concern. Restoration efforts within Mosquito Lagoon are focusing on reconstructing the three-dimensional reef habitats. Before effective protocols can be established, however, important questions about the sources of juvenile and adult oyster mortality must be answered. Potential causes of Crassostrea virginica mortality in the Indian River Lagoon system include sediment loads, competition, predation, and disease. My research focused on the interactions between oysters and the competitors that may affect the settlement, growth, and survival of Crassostrea virginica. The four objectives of my thesis research were to: 1) identify potential oyster competitors in Mosquito Lagoon, 2) determine if the sessile species recruiting to oyster shells have changed over time, 3) determine how the dominant competitors, barnacles, affect oyster settlement, growth and survival, and 4) determine if oyster or barnacle larvae are better able to settle in increased sediment and flow conditions that are associated with high levels of recreational boating. Lift nets were deployed within Mosquito Lagoon to determine available competing species. I collected species inventory data at six sites to determine the sessile invertebrate species (competitors) present on oyster reefs. Nets were deployed intertidally, just above mean low water, on living oyster reefs. One and a half liters of live and dead oysters were placed within the nets upon deployment. The nets were picked up monthly and surveyed for all fauna. Upon retrieval, all oysters within each net were brought back to the lab where all sessile organisms were immediately identified and returned to the lagoon. This survey began June 2004 and continued for one year. Shells from historic shell middens (up to 15,000 years old) were examined to determine if the sessile species settling on oyster reefs have changed over time. Similar species were found on both shells of historic and extant reefs. One notable exception was the appearance of Balanus amphitrite, an invasive barnacle, on the extant reefs. Balanus amphitrite is thought to have invaded Mosquito Lagoon approximately 100 years ago. This has resulted in a five fold increase in barnacle abundance per oyster shell. Balanus spp. were identified as important potential competitors and thus my research focused on spatial competition between C. virginica and native versus invasive barnacles of the area. Over 300 barnacles, including a native species, Balanus eburneus, and an invasive, Balanus amphitrite, have been counted on a single oyster shell. To determine how Balanus spp. affected settlement, growth, and survivorship of C. virginica, laboratory and field experiments were conducted in which densities of Balanus amphitrite and Balanus eburneus were manipulated. Density treatments included: no barnacles (control), low, medium, and high coverage of barnacles. Laboratory settlement trials with cultured oyster larvae were run in still water and flow (recirculating flume) using all barnacle density treatments. Additionally, all treatments with 7-day oyster spat were deployed in the field to follow oyster spat growth and survivorship. Settlement was counted by microscopy, and growth and survivorship were measured every 3 days for 4 weeks. Settlement of oysters was affected by barnacle presence only in flowing water. Still water trials showed no oyster preference related to any barnacle density or species. The presence of barnacles affected the growth and survivorship of oyster spat. However, there were no species specific differences. Studies suggest that recreational boating activities, especially boat wakes that cause sediment resuspension, may decrease recruitment and this may then provide an advantage to sessile competitors less affected by flow and sediment loads. To address these issues, replicated laboratory trials were run in a laboratory flume to quantify the effects of water motion (0, 5, 10 cm/s) and sediment loads (0, 8, 16 g/ml) on oyster recruitment and the recruitment of an important, relatively new competitor in the system, the barnacle Balanus amphitrite. If B. amphitrite settles in a wider variety of flow rates and sediment conditions, it may have a competitive advantage over the native oyster in this space-limited habitat. I found that high flow and sediment loads reduced larval settlement of C. virginica. Alternatively, settlement of cyprids of B. amphitrite did not differ among treatments. Thus, continuous boat traffic during settlement times should favor recruitment of the invasive barnacle Balanus amphitrite over the native oyster Crassostrea virginica. Determination of the competitive interactions of Crassostrea virginica in Mosquito Lagoon gives us important insights into the ecological conditions necessary for re-establishment of these oyster populations. Crassostrea virginica in Mosquito Lagoon was significantly impacted by barnacles; settlement, growth, and survivorship were all reduced by Balanus spp. This information will help resource managers in planning restoration techniques to minimize oyster and barnacle competitive interactions and increase Crassostrea virgininca success.

In addition to human activity, the Florida mangroves (Avicennia germinans, Laguncularia racemosa, and Rhizophora mangle) are being threatened by the invasive Brazilian pepper (Schinus terebinthifolius). This study was performed along a 261 km stretch of the east coast of Florida from Sebastian Inlet to the northern extent of mangroves, near St. Augustine. It entailed two parts. The first examined the phenology and leafing rates of the four species and attempted to find if there was a relationship between growth and latitude or temperature. Although a correlation between peaks in temperature and phenology was observed for all species, no leafing pattern could be discerned. In terms of mangrove growth for branch diameter, a logarithmic model (y=a + blog [Initial diameter]) best fitted the data for R. mangle and L. racemosa but neither latitude nor temperature appeared to be important. However, S. terebinthifolius' and A. germinans's branch diameter growth were best represented by a logarithmic model (y=a + blog [Initial diameter] + clog x2) that incorporated temperature and latitudinal respectively. In the second part, a simulation model was developed to focus on understanding the relationships between establishment and competition among the three mangrove species and the invading S. terebinthifolius. This model was run under various invasion and/or climate change scenarios to determine possible outcomes under global climate change with or without the presence of S. terebinthifolius. Conclusions were drawn that under all scenarios of invasion, other than sea level rise as part of global climate change, S. terebinthifolius would dominate the landscape if allowed to invade and establish in areas in which it is not currently present although the amount of this response is dependent on the S. terebinthifolius response curves.

Controlling and monitoring the BGL in post-op and intensive care patients in the hospital is also vital to their health. Currently the most reliable method of monitoring BGL is through an invasive procedure which monitors the amount of glucose in blood directly. A non-invasive glucose sensor would drastically improve the treatment of sensitive patients, and serve to improve the quality of diabetic patients’ lives. This glucose sensor is strongly based upon the mercury sensor developed by F.E. Hernandez and his colleagues. Glucose is used as a reducing agent to reduce mercury from Hg2+ to Hg0, which will form amalgams with the gold nanorods in solution. The change in aspect ratio of gold nanorods leads to a change in the UV-Visible spectrum of the solution. The blue shift seen was measured and correlated with the glucose concentration of the system. The system was then tested varying conditions such as pH, temperature, gold nanorod concentration, and mercury concentration. A preliminary study of the kinetics of the reaction was also done. The results showed a limit of detection of 1.58x10-13 and a linear dynamic range covering the concentrations of human tear glucose levels that are currently cited in the literature.

For example, the outcome of plant interactions can change depending on local environmental conditions such as nutrient availability, land management, or herbivory intensity. In particular, plant interactions may vary between facilitation and competition depending on ecological context, with facilitation expected to be prevalent under stressful conditions. I present the results of four studies that address different aspects of the community assemblage and dynamics emphasizing the synergistic effect of different processes. In the first, I investigated the importance of habitat isolation in determining species richness of wetlands with contrasting land use. The second describes an experiment to test the hypothesis that plant interactions with an unpalatable plant (Juncus effusus) would range from competition in ungrazed areas to facilitation in grazed areas and predicted that facilitative effects of Juncus would differ among functional groups of beneficiary species and be strongest when grazing was intense. In the third, I examine the community composition impacts of Juncus and predicted that Juncus would preserve functional diversity in grazed wetlands but that the effects of Juncus would vary along a grazing gradient. The fourth study investigated the relative importance of competition and nutrients in determining wetland invasion in two different land use types. Broadly, I demonstrate that the importance of different processes (habitat isolation, nutrient availability, competition/facilitation) to community composition is dependent on ecological conditions. This integrated view of community dynamics is interesting from a purely ecological perspective but also can be applied to understanding ecological problems such as exotic invasions and restoration of disturbed habitats.

Understanding the mechanisms underlying breast cancer progression remains urgent for developing effective treatment strategies to eliminate breast cancer mortality. Our recent studies have demonstrated that Kr(&)#252;ppel-like transcriptional factor 8 (KLF8) plays a critical role for breast cancer progression. Other studies have shown that Epithelial stromal interaction 1 (EPSTI1), a recently identified stromal fibroblast-induced gene in non-invasive breast cancer cells and epidermal growth factor receptor (EGFR) are highly overexpressed in aggressively invasive breast carcinomas including triple negative breast cancers. In this thesis project, we demonstrate high co-overexpression of KLF8 with EPSTI1 as well as EGFR in invasive breast cancer cells and patient tumors. We also show that KLF8 upregulates the expression of EPSTI1 by directly binding and activating the EPSTI1 gene promoter, and KLF8 upregulates the expression of EGFR not only by directly activating the EGFR gene promoter but also by preventing EGFR translation from microRNA141-dependent inhibition. Genetic, signaling and animal cancer model analyses indicate that downstream of KLF8, EPSTI1 promotes the tumor invasion and metastasis by activating NF-?B through binding valosin containing protein (VCP) and subsequent degradation of I?B?, whereas EGFR promotes tumor growth and metastasis via activation of ERK. Taken together, these data identify EPSTI1 and EGFR as novel KLF8 targets in breast cancer and suggest that KLF8 may be targeted for new effective treatment of breast cancer.

My manipulative experiments showed that the tested nonnative species influenced settlement, growth and survival of (<)i(>)C. virginica(<)/i(>) in unique ways. (<)i(>)Megabalanus coccopoma(<)/i(>) decreased the total number of settled oyster larvae, but did not influence larval preference or survival and growth of spat. (<)i(>)Perna viridis(<)/i(>) negatively influenced larval settlement and oyster larvae avoided settling on shells of (<)i(>)P. viridis(<)/i(>). (<)i(>)Mytella charruana(<)/i(>) had no influence on the total number of settled larvae but oyster larvae avoided settling on oyster shell with (<)i(>)M. charruana(<)/i(>) or on the mussel shells themselves. Furthermore, both nonnative mussels negatively affected the survival of juvenile oysters, but only (<)i(>)M. charruana(<)/i(>) reduced spat growth. These three nonnative species should be classified as invasive species because all had negative effects on the native oyster (<)i(>)C. virginica(<)/i(>).

Furthermore, the risk of complication from NPPV is much lower than MV, in terms of ventilator-associated pneumonia and sepsis. It is imperative for the nurse to understand the various indications, interfaces, and potential complications associated with NPPV use. In addition to treating acute exacerbations of COPD and cardiogenic pulmonary edema, NPPV has been used for prevention of reintubation, palliative care, and status asthmaticus. Furthermore, NPPV could be delivered through various interfaces, such as nasal, facial, and helmet. Each of these interfaces could eventually cause complications for the patient, such as skin ulceration and sepsis. However, there is limited amount of research available discussing the role of the nurse in caring for the patient with NPPV. There are no standardized guidelines established to assist the nurse in this care, in terms of interface selection, prevention of complications, and staffing patterns. Several recommendations are presented at the end of this thesis to guide future nursing research, education, and clinical practice, such as exploring the role of oral care and education for NPPV patients.

Show moreEvaluating relative species competitive strength is a central question in community ecology, with strong implications for invasion ecology. Models assessing invader success consider three components: distribution, abundance and per-capita impact. However, relative strength and interactions among these factors remain unclear when applying to specific invasion scenarios. We hypothesized that performance of native and non-native species will vary as a function of direct and indirect effects at different abundances and scales. We conducted a replacement experiment between two dominant grasses in subtropical grasslands (the native Axonopus fissifolius and the non-native Paspalum notatum) in central Florida, USA. Thirty fenced plots (1 m x 3 m each) representing a gradient (15 levels) of increasing non-native groundcover and decreasing native groundcover were set up in November 2017. We transplanted individuals of these two species in subplots (12 subplots and 36 transplants per plot; 1080 plants in total) in a 2*2 factorial design (mixed /single focal species * 2/4 transplants per subplot). Leaf length/number and plant biomass were evaluated at the beginning and end of the experiment along with plot species composition and soil nutrients. Over 92% of transplants of each grass species survived until harvest (11 months). There were significant differences in leaf length, leaf number and plant biomass between conspecific/allospecific subplots. Both P. notatum and A. fissifolius performed better when transplanted in non-native P. notatum subplots. There were also interactions between conspecific/allospecific subplot treatment (direct effects) and the gradient of increasing Paspalum notatum /decreasing Axonopus fissifolius groundcover (indirect effects) treatments. Increasing P. notatum in the whole plot made environments more favorable for both grass species. Effects were consistent for leaf length/number and biomass of the two focal species. More comprehensive evaluation on indirect effects need to be considered when examining competition between non-native species and native species.

A standardized and effective antibiotic therapy against MAP and/or AIEC is needed for better treatment. Three antibiotic drugs (-) Clarithromycin (CLA), Rifabutin (RIF), and Clofazimine (CLO) have been used to treat CD patients suspected with MAP infection. However, the outcome has been controversial. The treatment dosage is high, the duration is long, and the reported drug side effects resulted in patient non-compliance; therefore, a lower and effective drug dosage is needed. In this study, we developed two aims 1) to evaluate RHB 104, a drug formula comprised of low dosages of CLA, RIF, and CLO, against clinical MAP strains in-vitro using fluorescence quenching method, and 2) to develop a fluorescence in-situ hybridization method to detect both MAP and AIEC simultaneously in intestinal tissues of CD patients. A total of 16 clinical MAP strains and 19 non-MAP strains were tested against varied concentrations of RHB 104, CLA, RIF, and CLO. Although the MIC for all drugs ranged between 0.5-20 ?g/ml, the MIC for RHB 104 was significantly lower against most MAP strains. The effect of RHB 104 against MAP was bactericidal. Unlike RHB-104 formula, CLA, CLO, and RIF dosage similar to those in RHB-104 did not inhibit MAP growth when trialed individually and in dual-drug combinations. The data illustrated the presence of synergistic anti-MAP activity of low dosage of the three antibiotics in RHB-104. We also developed a rapid and sensitive multicolor in-situ hybridization technique that can detect MAP and AIEC using tagged-oligonucleotide probes. Non-pathogenic Escherichia coli (npEC) was used as a control for the study. Specifically, cultured MAP and npEC were fixed and hybridized with MAP488 and EC647 probes, respectively. Confocal laser scanning microscope (CLSM) revealed specific signals at 488nm for MAP and 647nm for npEC, indicating probe binding to each bacteria. This was confirmed with hybridization of MAP with EC647 and npEC with MAP488 resulting in absence of signals. Intestinal tissue samples from 9 CD patients were then analyzed using our technique. Preliminary data indicated positive results in 6/6 samples for MAP, 6/6 for npEC, 3/3 for AIEC, and 2/2 for both MAP and AIEC with MAP being more dominant. This protocol shortened the FISH procedure from multiple days to short-hours. The protocol allows the investigation of more than one pathogen simultaneously in the same clinical sample. A quantitative measurement of the signals is needed.

Show moreThe Nile monitor lizard [Varanus niloticus (Linnaeus, 1766)] is a generalist carnivore, native to Sub-Saharan Africa and the Nile River but now established in North America as a result of the pet trade. Once introduced, they are a potential invasive threat to native wildlife. Here, I create ensemble species distribution models (SDMs) to predict the global distribution of this generalist carnivore given current and future climate conditions. I then quantify the monitor's potential effects on 85 food webs representing (>)900 different species within the projected regions based on stomach content data. Climate, vegetation, and elevation data are used for 507 georeferenced observation points from the Nile monitor's native range to produce current and future (2070) ensemble SDMs. Explanatory variables are evaluated as ten alternative models organized in three subsets according to model assumptions. The true skill statistic (TSS), sensitivity, and specificity were used to assess model performance, and the best subset was averaged to represent an ensemble model. Food web impacts after the generalist predator's addition are determined by changes in nine metrics of food web structure. The most predictive (TSS scores ?0.87) ensemble SDM was based on the MARS and FDA algorithms using elevation and climate for current and future conditions. This model shows that, if introduced, Nile monitors will likely spread into many regions in the Americas, the Caribbean, Madagascar, Southeast Asia, and Australia. Assuming unabated carbon emissions by 2070, climate change will enhance that potential range. Adding Nile monitors to food webs generally increases overall trophic links, connectance, link density, and fraction of intermediate taxa, with decreases in the fraction of top and basal taxa. These results are consistent with a generalist predator that affects many species and is likely to affect food web stability. The potential Nile monitor range is vast and encompasses multiple biodiversity hotspots. Given many strong food web interactions by this generalist predator, vulnerable regions should actively prohibit/regulate Nile monitors as pets, enforce those restrictions, and promote exotic pet amnesty programs. Southern US states should especially act soon to prevent spread of the Nile monitor to the Neotropics from its current introduced population in Florida and as released pets.

Invasive methods of ICP monitoring include lumbar puncture manometry, which requires high precision, is costly, and can lead to complications. Non-invasive monitoring of ICP using tympanic membrane pulse (TMp) measurement can provide an alternative monitoring method that avoids such complications. In the current study, a piezo based sensor was designed, constructed and used to acquire TMp signals. The results showed that tympanic membrane waveform changed in morphology and amplitude with increased ICP, which was induced by changing subject position using a tilt table. In addition, the results suggest that TMp are affected by breathing, which has small effects on ICP. The newly developed piezo based brain stethoscope may be a way to monitor patients with increased intracranial pressure thus avoiding invasive ICP monitoring and reducing associated risk and cost.
