Show moreStomata are cellular structures that control water loss and gas exchange through the plant epidermis. Stomata arise from special stem cells called meristemoids through a series of programmed asymmetric divisions that are controlled by cell signaling, or via multitude of regulatory pathways and intercellular communication between epidermal cells. In Arabidopsis thaliana, stomata are spaced non-randomly in the epidermis by cell-cell signaling of the receptor-like protein TOO MANY MOUTHS (TMM) as well as other proteins. Point mutation of the TMM gene prevents the development of stomata in some tissues like inflorescence stems. Investigation of tmm mutant stems showed that self-renewing stem cell-like precursors form by dividing asymmetrically but fail to form stomata. This is further supported by molecular markers of stomatal cell fate that show stomatal precursors form but do not differentiate as stomata. Therefore, TMM signaling is likely required to control expression of genes that are essential for the formation of stomata in stems. As a second approach, gene expression profiling was used to identify candidate genes involved in stomatal biogenesis. Differentially expressed genes were categorized by gene ontology and analyzed for statistically overrepresented classes to gain insight into functional processes. Comparison of stem expression data with previously published microarray data was used to narrow the list to genes involved in stomatal patterning. Mutants in these target genes have been obtained and phenotypic analysis revealed new stomatal regulators. Comparison of epidermal cells of the stem tip region from wild-type and tmm revealed that there are significantly more meristemoids formed in tmm stems compared to wild-type stems. In addition, the orientation of meristemoids formed in wild-type stems was random with respect to stem polarity and followed a spiral pattern of asymmetric divisions similar to leaves. This showed that stomatal patterning in dicots does not follow orientation in asymmetric cell division for spacing the adjacent stomata like monocots.

Show moreThis master's thesis project focuses on the life and music of Liza Minnelli, the research of which culminated in the development of a one-woman performance piece and monograph. Using song, dance, and dramatic dialogue, this presentation reveals the many aspects of Liza's heart and soul, her relationship with her famous mother, and her own personal struggles. Through the eyes of Liza, this piece will explore the duality of her life--the underlining turmoil that fluctuated between her personal life and professional career. My methodology includes studying biographies, articles, and interviews written about Liza Minnelli, as well as watching films and performances of her work. The performance piece is a compilation of my research. I will also explore the relationship and influence that Liza's famous mother had on her, and see if other adversaries in her personal life affected her choices as a singer, actress, and entertainer. People have often told me I look like Liza Minnelli. Walking down Lexington and 47th Street was one of the many times; I was approached by excited Liza Minnelli fans wanting an autograph and snapshot of me. The reaction I received in letting them know I was in the same business was just a precursor to daydreaming of one day portraying Liza on stage. I always loved Judy Garland's and Liza's musicals and movies. They inspired me at a young age. Never once did I try to emulate Liza's style or copy her personality, because like her, I am determined to be my own "me." Ultimately, I would like this to be the beginning of something that I can continue to develop and workshop with the intent to produce for other professional theaters.

In 1985, Christopher Durang created a master work titled The Marriage of Bette and Boo which was described by The New Yorker as a "brimming cornucopia of brilliant lines." Frank Rich of the New York Times called it "so speedy and chipper that it could almost be mistaken for a Bob Fosse musical." Douglas Watt of the New York Daily News referred to it as a "farcical study of a disastrous marriage, marked by still births, insanity in the family and divorce." Show moreIn 1985, Christopher Durang created a master work titled The Marriage of Bette and Boo which was described by The New Yorker as a "brimming cornucopia of brilliant lines." Frank Rich of the New York Times called it "so speedy and chipper that it could almost be mistaken for a Bob Fosse musical." Douglas Watt of the New York Daily News referred to it as a "farcical study of a disastrous marriage, marked by still births, insanity in the family and divorce." These critical responses indicate the necessity to maintain the complexity of the character of Soot without allowing her to fall into the easy stereotypes that trap many artists. The play's focus on alcoholism, spousal abuse, and cancer make it difficult to reconcile the very serious circumstances with the overtly comical reactions created in the play. My challenge in portraying the character of Soot is to resolve these issues. I focused on the teaching of Konstantin Stanislavski as interpreted by Sonia Moore of the American Center for Stanislavski Theatre Art to navigate this challenge. Ms. Moore asserts: "Only after the actor has studied the play, the events, and the given circumstances will he be able to select the actions which will involve his emotions and other inner experiences." With this in mind, I began by studying the script and the playwright for clues about the given circumstances within the world of the play. "You're the dumbest white woman alive" is a line from the play used to describe Soot. This line of dialogue has been used in the past to justify stereotypical performances of this character which mask the complexity of a woman who has been seriously hurt by the man she loves. These types of simplified characterizations are flat and do not take into account the inner and outer forces that make Soot who she is. Olympia Dukakis played Soot at the Public/Newman Theatre in New York City in an Obie award-winning performance. She is quoted as saying, "This is a very forgiving play." Durang later agreed with this assessment saying, "I remember thinking that that sounded right." He further added that The Marriage of Bette and Boo is "based on my parents, it's more emotionally close to me than some of my more surreal plays
I like the balance of the comic and the sad. It should play as funny, but you should care about the characters and feel sad for them." Durang also confides that his own father was an alcoholic and that "in life, my mother lost three [children in childbirth]." Since the play, however humorous, is based on real events in the life of the playwright and his family, it is reasonable to search for playable and understandable motives for the character's action through research of real life given circumstances. To this end, I have been researching the behavior rationalizations inherent in the social interactions of alcoholic families. This has led me to discover a possible explanation for Soot's unusual responses to unpleasant comments and situations. The eGetgoing Online Addiction Treatment Alcohol and Drug Rehab Counseling web page explains that family members often choose enabling behavior to cope with the destructive choices of the alcoholic. Enablers "may have their own system of denial that is fed by the lies and deceptions." Further, it states, "We can think of denial as a way of telling the truth about a small part of reality as if it were all of reality." In this way, Soot is employing a rational tactic to cope with an irrational existence. I believe further research will also identify denial associated with personal tragedy. These understandable and human connections will provide me with the basis of my beginning explorations of this achingly beautiful and sympathetic woman because, as Sonia Moore states, "A person's psychological and physical behavior is subject to the external influence of his environment." Soot's environment includes the time period of the piece and is complicated by the memory play nature of the script, but it adds context for Soot's behavior as a subservient 1950's housewife who feels as if she has very little control over her own world. According to Elaine Tyler May in her book Homeward Bound, "If a woman was not satisfied being a homemaker she was supposed to keep that information hush-hush and go about her daily activities as though nothing was wrong." This information provides further justification and adds strength to the logic of Soot's actions as she deals with the dysfunctional world of her family. The work I have done researching Soot's motivations appears to be relevant to the execution of the play. I may now begin to apply that research to the creation of Soot. The role of Soot provides a wonderful opportunity to develop and grow as a performer. A thesis role needs to supply the MFA candidate with significant challenges. This role has already provided huge opportunities to connect my research skills with my stage work. I believe the role of Soot will force me to push myself and apply all the skills and training I have received during my studies in order to do justice to the richness of the character.

In the middle of the collection (")The Mad Scientist Sleeps(") and (")Through Milk and Oil(") surround (")Insomnia and Autocannibalism,(") reaching the core of the speaker's identity throughout the collection, imagined, present, or past. The collection, like its namesake the ouroboros, ends in the same place it begins. This cyclical motion through the collection seeks to bring the varying voices throughout into a complete, if conflicted whole.

Within this collection, festivals are illustrated as more than just spectacular stages and bright lights(-)they're depicted as fascinating, budding utopias that encourage creativity, generosity, and positivity from attendees who abandon inhibitions, and oftentimes logic, in the name of fleeting freedom from the routine of their (")real(") lives. The narrator strives to live a fulfilled life(-)what many might call a well-lived life, if not a privileged life(-)but she struggles to identify her life as meaningful as she works to disentangle the falsities of her (")real(") life as typically defined by society, a corporate, desk life in between festivals, and her electric life, an actualized but less publicly-accepted life at festivals. She repeatedly contemplates her relationship with art, and whether or not art offers a sort of immortality to those who pursue it. As a festival-goer, she finds that the art of music takes her away from her own art, writing, but her writing is about the festivals, so a love/hate relationship grows with the festivals over time. Many of these essays, such as (")In a Tent, a Home,(") (")Rebecca,(") (")We Left Town,(") and (")I Don't Wanna Wear No Shoes,(") ruminate on how dislocation and travel can be fulfilling occasions for further ontological inquiry. And the world, despite its familiarity, can be a hostile place. The characters in this collection learn that families are a fragile lot, that every desire contains a paradox, that the Road of Life can seemingly be grasped by the horns, but that the future twists and turns, yet never escapes the past. And it is the past that haunts these characters' lives. One word, one act, impacts a lifetime. In A Little Slice of the Moon, Khalid traces the devastation of his 'new' life and his alienation to everything around him back to a youthful error. In The Thousand Trees Orchard, the arrival of Mahjabeen, Laddo's deranged and possibly dangerous sister, teaches Laddo the difference between fleeing the past and embracing it. In Dead Woman's Pass, Priya tries to outrun her malevolent qismet, and in doing so, almost loses herself as well. Isolation, physical or emotional, is a primary element in many of these characters' lives. Whether the isolation is self-imposed or results from circumstances beyond their control, these characters realize that where they are matters less than what they've done. They learn that confronting themselves--who they are, who they were--is the only way to break free from the past and make peace with themselves and with the world around them.

It was shot in Ecuador with non-professional actors and a minimalistic production style. These characters are often obsessive and do not trust one another. They think life is funny, and discover that love is funny, and yeah, sex can be funny too. They don't feel the right things when they're supposed to. They find love, and lose love. They find hope, and lose hope. They escape sometimes, but more often are unable to go anywhere. These stories consider relationships through the disconnection between reality and fantasy, exploring how the lines between illusion and actuality can become blurred. A young boy fantasizes about running on the wind; teenagers pretend to be werewolves; twenty-somethings obsess about potential love affairs, dreams, and the possibility of escape. There is a driving curiosity behind these characters, a desire to figure one another out(-)a desire to learn the other's secret. Trade secrets are insider information after all, and must be earned. These characters are all earning the right to hold their own trade secrets and, when the time is right, sharing that information with whomever is willing to listen.

Show moreABSTRACT In the fall of 2007, I was cast in the University of Central Florida's production of The Piano Lesson. My thesis will examine my performance in the role of Grace, as well as understudying the role of Berniece under the direction of Professor Belinda Boyd. In addition to the performance components, my thesis materials will include historical and cultural character research and a reflective journal documenting my rehearsal and production process. My character research and journal will address questions about the characters "choices" and the relationship of their environment to views of personal responsibility and obligations in the play. I will examine the characters' relationships and situations in the play, investigating some of the decisions that these characters make in response to their cultural and social landscapes. The second part of my thesis will include research on three additional female characters from August Wilson's dramatic canon: Aunt Ester Tyler from Gem of the Ocean, Ma Rainey from Ma Rainey's Black Bottom, and Rose from Fences, which will inform my character explorations of Grace and Berniece, and the choices that I make during the rehearsal process in developing their journeys. By reading other plays by Wilson, I endeavor to deepen my understanding of the struggles of African-American women in the twentieth century, and explore how the social and economic status of black women was affected by America's changing political and social climate over several decades. In addition, I will document how my visceral experience of performing the roles of Grace and Berniece relates to my intellectual process of exploring the journeys of Aunt Ester Tyler, Ma Rainey, and Rose.

Show moreQuantitative studies of the presidential use of executive orders have attempted to determine whether presidents are more prone to resort to unilateral action when faced with legislative opposition. To date, the results have been mixed however, with studies demonstrating that the type of executive order is an important factor in understanding the conditions under which presidents will resort to unilateral action. Despite this advancement in theory, there has been little consensus regarding the actual conditions under which presidents will issue the different types of executive orders that have been identified in the literature.This thesis addresses this puzzle through an empirical analysis that engages the "Two Presidencies Thesis," which argues that presidential decision-making, action and success is conditioned by policy area (foreign and domestic) and executive order type (major, routine, or symbolic). An original dataset was constructed by coding all executive orders issued between 1953 and 2008 as related to either foreign or domestic policy. Thus, an analysis is undertaken of major executive orders, minor executive orders, foreign policy-based executive orders, domestic policy-based executive orders, and major and minor categories of each policy area.A multivariate analysis is completed using negative binomial regression given that the dependent variables are overdispersed count variables. The effects of divided government and ideological distance are the primary independent variables examined. The ideological distance variable consists of the absolute distance between the president's ideology and the ideology of the median member of the Senate. Various other control variables are included, including presidential party, election year, and approval ratings. The findings indicate that executive order type does matter in predicting presidential use of executive orders and that the prevailing political climate does influence the president's use of executive orders.

The Musical is a comedic parody based on Herman Melville's 1955 classic novel Moby Dick about a madman who seeks revenge on the great white whale that crippled his body and consumed his spirit. The thesis role I have chosen is "Starbuck", the ship's first mate. If this were a dramatic telling of the classic tale, my role would be considered an absurdity for the sure-known fact that Starbuck is a male character. However, since Moby Dick! The Musical is a spoof that features a play within a play, many, if not most, of the roles are being played by women (teenage school-girls to be exact). These roles are known as "trouser roles," and this tradition stems back to the 17th Century. The term refers to a male character sung by a woman (mezzo). It is also referred to as a "breeches part" or in Italian, "travesti". This will be my first trouser role experience. Before today, I hadn't given the concept much thought in relation to musical theatre. These roles generally live in works ranging from Shakespeare to early operetta, and most important, Opera. This thesis role will allow me to log a personal experience in journal form and experience those challenges and rewards that transpire from a live performance. My research will include the history of the "trouser role," including famous performers, specific roles in shows, and the effect it has had on audiences over the years. My main concern, however, is when, where, why, and how the concept made its transition to musicals. It will also be interesting to see what genres these roles are generally written for. Are they all parodies like Moby Dick! The Musical or are there a few dramas thrown into the mix? There will surely be a long list of shows that include the "lady in drag". When all is said and done, I will have a wealth of information in an educational thesis that will prove the significance of an ever-transforming concept.

I will take each episode and divide it into the following sections: given circumstances, what is said about the character by the playwright, by others, or by herself, objectives, tactics, vocal traits, and physical traits. The third phase will include a written journal of my experiences as an actor as they occurred during the rehearsals and performances. The fourth and final phase will include a self-analysis of the performance. I will reflect on my abilities in synthesizing the research and character analysis found in phase one and two into the actual performance. In addition, Committee Chair, Mark Brotherton, and my thesis Committee Members, Kate Ingram and Vanduyn Wood will also give written responses. The performances will be held February 14-17, and 21-24, 2007 in the University of Central Florida's Black Box Theatre. Dr. Julia Listengarten will direct the performance.

Without the financial and scheduling constraints of enlisting a large crew, the film was allowed to thrive with a spontaneous and ongoing shooting schedule, controlled entirely by one person.

This thesis is useful, important, and unique as it focuses on the stories of a variety of characters, mostly women and children, and displays the beauty and fearsome power of nature as the characters strive to achieve their goals. In today's political and social climate, women, children, and nature are often taken for granted, underestimated, and even forgotten about. Here, they are anything but forgotten. Women join together to fight trauma, children stand together and face some of today's worst natural disasters, and nature is portrayed as a source of magic. This thesis gives these characters a voice and shines a spotlight on their importance to the world and society as a whole.

Show moreMy thesis, a collection of personal essays, explores my parents' affinity towards their native Colombia and how this connection to their homeland, through their faith and their customs, affected my definition of self. When I think about my parents' emigration from Colombia to the States, I picture the illustrations in the Bible I had as a child: the couple running from Sodom and Gomorra, running away from the place they had always known and holding on to each other. My parents, like the couple in the Bible, were in the middle of nowhere when they first set foot on the cold, concrete streets of New York City. In the Bible, the man knew he was in a better place, the cities left behind him becoming more and more of a distant memory. The next picture showed a statue of salt in the shape of the woman. The woman had turned back. Shortly after they married in Colombia, my mother looked forward to a future in another country. She urged my father to seek a better life for them in the United States. My father was the one who couldn't help but look behind him, despite the consequences. The thesis chapters explore such issues as the consequences of leaving home; the impact of my father's incarceration upon his Catholic faith and upon the family; how travel to Colombia with my parents revealed new aspects of their personalities and beliefs; and my own efforts to understand and meditate upon my multicultural heritage and surroundings.

Show moreWhat Instructional Modification Implementations Bring Adult Education Learners to Success and Retention? Analysis of an Online Adult Basic Education Program. Adult learners enrolled in my online Adult Basic Education program showed low completion rates for the course. This study's purpose was to discover what instructional modifications would bring these learners to a successful outcome in the program. I implemented the following instructional modifications: asynchronous email communication, weekly progress reports emailed to each participant and a minimum requirement of 10 assignment completions on a weekly basis. Teacher-student interaction increased because of these implementations. I reviewed literature that discussed distance education, adult learners and teacher-student interaction to discover strategies that would improve student achievement in an online course. After reviewing the literature, I determined that there were three missing elements in my program: structured communication between the teacher and student (Palloff & Pratt, 1999), standards for online coursework completion (Comings et al., 1999) and a system to inform students of their progress in the program (Comings et al., 1999). I found that teacher-student asynchronous communication, a weekly requirement of 10 completed lessons and weekly progress reports of student accomplishments encouraged students to complete coursework on a regular basis. Students were conscientious, taking more responsibility for their learning. Students had a higher rate of coursework completions during the research study period, with performance increasing 73%.
