My work deals with the subconscious pairing of colors from an instinctively chosen color palette into molded forms. My emotions drive these pairings. As I visualize these different emotions, I ask the viewer to formulate their own perceptions and connections (both physical and emotional) in relation to my own. This allows the viewer to explore the emotional twists and turns of my imagination by their own unique associations in correspondence. Painting allows me to visualize the inner depth of my imagination. My work becomes concrete through the subconscious pairing of colors and shapes and evolves with viewer interaction.

In it I describe and analyze multiple influences tied to the progression and change in my studio practice. I began the process of my work with images and subjects from my home state of Florida, frequently juxtaposing the wildlife and humans, now I see this pattern as a byproduct of a studio practice functioning as a introspective reflection of what I experience, the things that I understand and the things that I don't. I deconstruct elements of figures and landscape, removing most information but what is necessary to retain symbolic context, and allow physical windows into the past formal states of the work, exposing the audience to different periods of time and hinting at information now hidden under the finished image.

The artist possesses a creative impulse finding satisfaction in making artwork that expresses a love of nature as part of a larger worldview. If done well, the persuasive power of such art may be enormous. Comprised of oil paintings and written work, this thesis establishes a way of approaching both landscape painting and the natural environment. Literature pertaining to the contributions of landscape artist Frederic Church, varying aesthetic theories, nature writings, and selected contemporary artists are discussed. The focus then turns to particular landscape elements, introducing the artwork created for the thesis. The thesis concludes with the artist's purpose statement.

Found and appropriated furniture and fixtures from the actual rooms are arranged within the replica as objects of art to familiarize the public with conditions of poverty.During my exhibition, the public is encouraged to participate by entering and exploring the replica of the motel room. The participant experiences authentic objects, sounds and smells of Daytona Beach motel rooms that have been inhabited by victims of transient poverty.?

I believe there is more to say in a landscape with the absence of the figure, allowing the dialogue to be read and interpreted by a larger audience. I am particularly satisfied with White Knuckles, shown in figure 3, because I collide reason with imagination, thus contradicting the context and interpretation of the subject matter. In White Knuckles, I deal with formal elements such as composition, atmosphere, lightness and darkness as well as nuances of color. I have also considered the emotive impact the painting could reflect to the viewer, specifically feelings of tension and unease. The placement of the viewer outside the picture plane was carefully considered to suggest the audience is a participant in the suggested narrative. I often strive to create an ambiguous moment, reflecting feelings of uncertainty and apprehension. Like White Knuckles, my body of work employs unexpected narratives to reveal some of the uncomfortable truths of our human experience. I am interested in exploring the relationship between the mundane and the abnormal in the paintings, a feeling that could be described as a (")suburban nightmare.(")

Laser-induced breakdown spectroscopy (LIBS) was the central instrument utilized for analysis although scanning electron microscopy / energy dispersive x-ray spectroscopy (SEM/EDS) and Fourier transform infrared spectroscopy attenuated total reflection (FTIR-ATR) analyses were also performed. Two separate methods of LIBS analysis of samples were used: a cross sectional analysis and a drill down analysis. SEM/EDS analysis focused on the cross section while FTIR-ATR analysis concentrated on the clearcoat layer. Several different data/statistical analyses were evaluated including principal components analysis (PCA), two tailed t-tests based on several different metrics (Hit Quality Index (HQI), Pearson's correlation and Sorenson index), multivariate analysis of variance and receiver operating characteristic (ROC) curves. Full spectrum data analysis from LIBS spectra resulted in 99.7% discrimination between different sample comparisons and 12% between same sample comparisons based on HQI and t-tests. Peak analysis of LIBS spectra resulted in 87.5% discrimination between different sample comparisons and 5% between same sample comparisons based on MANOVA. When combining the results of the FTIR-ATR and SEM/EDS analyses, 88% of the samples could be discriminated.

This production premiered on the University of Central Florida's Mainstage Theatre 29 March 2007 and ran for fifteen performances, closing on 15 April 2007. This thesis includes my research and analysis of the published script and a production journal which documents my process for our specific production. In this document, I illustrate how my communication and meetings with the director and members of the design team moved from initial conversations into the research, development, and construction processes in order to create the realized production. I explain the challenges and obstacles I faced in the development and construction stages and how my communication with members of the design and production staff led to the final design. Throughout this document, I present figures displaying the preliminary sketches, renderings, construction process, and production photographs that illustrate how the production moved from an idea to a reality. I also present a self analysis of the process and final product through personal reflection and outside response.

Show moreProstate cancer is the most common cancer in men in the western world. Although early stage prostate cancer is treatable late stage, more specifically, metastatic and drug resistant prostate cancers are mostly incurable. The failure of current treatments obligates the research community to explore novel areas in prostate cancer biology and find better therapeutic targets. Emerging evidences show that non-coding RNAs specifically long non-coding RNAs (lncRNAs) play regulatory roles in various cellular processes and are frequently dysregulated in cancer including prostate cancer. These aberrantly expressed lncRNAs mostly with unexplored genetic information may drive cancer progression. Previous studies done in our laboratory showed a tumor suppressor role of a cluster of small non-coding RNAs or microRNA (miRNA) miR-17-92a in PC-3 prostate cancer cells. To learn the underlying mechanism, transcriptome analysis with or without expression of miR-17-92a was conducted in our laboratory. RNA-sequencing data analysis identified reduced expression of a set of lncRNAs and oncogenes, and up regulation of several tumor suppressor genes upon expression of miR-17-92a cluster miRNAs. One of the down regulated intergenic lncRNAs, PAINT (Prostate Cancer Associated Intergenic Non-coding Transcript) (LINC00888), was selected for determining its functional role in prostate cancer. TCGA and GEO profiles analyses revealed up regulation of PAINT in prostate tumors with higher Gleason Scores, in highly aggressive metastatic prostate cancer cell lines, and upon androgen deprivation therapy of prostate cancer cells. This observation was supported by our studies on expression analysis of PAINT in prostate tumor tissues using RNA in-situ hybridization in tissue microarrays (TMA) containing tissues from different stages of prostate cancer and normal prostate tissues, which showed higher expression of PAINT in prostate cancer tissues compared to normal tissues. Furthermore, late stage (stage III and stage IV) prostate tumors showed significant overexpression of PAINT compared to early stage (stage II) prostate cancer tissues. We examined the functional relevance of PAINT in promoting tumor progression next using different prostate cancer cell lines. Silencing of PAINT using siRNAs showed decreased cell proliferation, reduced S-phase progression and activation of pro-apoptotic proteins PARP and Caspase-3. Silencing of PAINT also showed decreased cell migration and increased expression of the epithelial marker, E-cadherin while reduced expression of mesenchymal markers Slug and Vimentin. Ectopic expression of PAINT reversed the effects observed upon silencing of PAINT. Increased cell proliferation, cell cycle progression and cell migration were noted in prostate cancer cells overexpressing PAINT. Additionally, cancer promoting phenotype such as larger colony formation and higher expression of mesenchymal marker Slug, was detected upon overexpression of PAINT. Our study also determined the therapeutic benefit of inhibition of expression showing an increased sensitivity of metastatic prostate cancer cells to the chemotherapeutic agent docetaxel (DTX) and selective Aurora kinase inhibitor VX-680. Taken together, our study establishes an oncogenic function of PAINT, its clinical relevance as a marker for advanced stage prostate cancer and its potential as a therapeutic target for metastatic prostate cancer.

Show moreThis thesis examines the concepts and ideas that pervade my body of work in painting. These concepts include immersion and scale, interactions in space, harmony and disunity, flat and dimensional space and perhaps most importantly, ambiguity. I intentionally use these formal devices to create immersive environments that appear as a familiar but skewed version of reality. By forcing together disparate languages of visual representation into the theoretical space of a painting, I strive to expose some of the humor and decay of our social institutions. In using the format of collage, my paintings have the opportunity to incorporate a wide array of iconography and imagery, while adding commentary through their juxtapositions. This thesis further explores the notion of oversaturation of images in media and how it has resulted in a clashing of imagery in the public space, akin to the format of collage in art-making. By using more easily readable or accessible iconography like cartoons or digital images to draw viewers in to the work on a visceral level, I discuss how the audience becomes witness to something sinister or something in the process of decay. Since I do not fully understand my own position as an artist and consumer of images, painting serves as a way to examine and question my relationship to society and culture as a whole. The artworks are inevitably questions in themselves: Who is to blame? What does it mean to be American? What is my duty as an artist?
