Stalled ribosomes have profound effects on a cell such as altered mRNA abundance, decreased ribosome availability, and an imbalanced proteome. The absence of elongation factor P (EF-P), a universally conserved transpeptidation enhancer, presents an extreme example of this scenario, wherein ribosomes accumulate disproportionately onto messages that are more slowly translated and cell growth becomes notably impaired. We discovered that faster-growing cells arise spontaneously in ?efp cultures, suggesting that translation defects could be circumvented by mutating other genes. This thesis presents a genetic and biochemical analysis of a mechanism ?efp cells employ to overcome translation stress. Using a dual luciferase reporter system, we found that transpeptidation remained hindered in the faster growing ?efp cells. Whole genome sequencing of several fast-growing strains revealed mutations in a poorly characterized RNA helicase called HrpA. We determined that deletion of hrpA, or mutations at several conserved residues critical for HrpA's function, was sufficient to improve the fitness of ?efp cells. HrpA is a DEAH-box RNA helicase and represents a large class of enigmatic proteins that use ATP to restructure cellular RNAs; however, it's direct function in cellular physiology has yet to be clearly demonstrated. Several HrpA mutants were engineered to interrogate the molecular mechanism of HrpA and how its function impairs ?efp cells. Complementation in ?efp ?hrpA cells showed that a number of these mutants were unable to restore sickness, suggesting they were defective in key aspects of RNA processing. It was discovered that wild-type HrpA is associated with actively translating ribosomes and several of the inactive HrpA mutants impose substantial deleterious effects on translation and ribosome production. In sum, the work presented here describes a mechanism by which cells overcome translation stress involving a novel genetic and biochemical relationship between EF-P and HrpA.

A bilingual advantage has been observed in various experimental studies, but also has not been observed in many other studies. A bilingual advantage has been shown in tasks using selective attention. These tasks study the effects of language interference, where two types of interference are observed: interlingual (between-languages) and intralingual (within one language). This study examined language interference in Spanish-English bilinguals, using an auditory-visual simultaneous translation experimental setup. 16 college English monolinguals and 17 college Spanish-English bilinguals were tested. The task was to ignore the word in the headphones and to translate/repeat the word on the screen into English. Distractor words went to either the right, left, or both headphone ears. Subjects were given 72 words to translate, words were randomized, and ear of the distractor word was randomized. The monolingual group was not affected by any independent variables tested except screen word length. Bilinguals did worse when the word and audio were in Spanish, and when the word and audio were different words. No ear advantage was observed. Proficiency levels and first language had no effects on bilingual performance. More intralingual interference was observed for bilinguals only, no significant interference occurred for monolinguals. A slight bilingual advantage was found but not fully, because of the high load of the task and introduction of another language. In conclusion, bilinguals did not have a cognitive advantage in this experiment setup.

Show moreThis paper investigates the most effective ways of handling cultural differences in the Japanese-to-English game localization process. The thesis advocates for applying the Skopos theory of translation to game localization; analyzes how topics such as social issues, humor, fan translation, transcreation, and censorship have been handled in the past; and explores how international players react to developers' localization choices. It also includes interviews with three Japanese-to-English translators who have worked with major Japanese game companies to gain insight into how the industry operates today. Through the deconstruction of different aspects of Japanese-to-English localization, this analysis aims to help the game industry better fine-tune Japanese media to Western audiences while still sharing valuable aspects of Japanese culture. The thesis concludes that if Japanese game companies work to improve the localization process by considering more diverse international perspectives, hiring native speakers as translators, and approaching the English script as a creative endeavor in itself, they will be able to both broaden the minds of their global audiences and more effectively capitalize on the worldwide fervor for Japanese video games.

Show moreIn this qualitative study, I examine the utilization of electronic publication and electronic writing systems to provide new possibilities for the translation of French Caribbean literary texts. Using Simone Schwarz-Bart's 1972 novel "Pluie et vent sur T(&)#233;lum(&)#233;e Miracle" specifically for analysis and exploration, I investigate the potential of digital technology to aid in the production of literary translations that are mindful not only of the dynamics of language, but of French Caribbean women's discourse as well. Since the cultural turn of translation studies, translators need not only be bilingual but bicultural as well, having a discerning knowledge and familiarity of the culture that they render. Cultural translation scholars, therefore, have argued that translators should make the reasons for their translation choices known through annotations, prefaces, introductions, or footnotes. Advancing this established claim through critical and theoretical analysis and the construction of hypermediated textual translation samples from Pluie et Vent, I argue that translators can make their choices known by utilizing digital writing and hypermedia tools, such as TEI-conformant XML, and using them for computer assisted translation (CAT) and electronic publication. By moving a new translation of Schwarz-Bart's text to a digital space, translators have more options in how they present their renderings including what information to include for better textual interpretation and analysis. The role, thus, of the translator has expanded. This person is not just a translator of language and culture, but an editor who provides scholarly information for critical interpretation. She is also a programmer who is skilled in new media writing and editing tools and uses those tools rhetorically to invent new methods for the electronic translation of literature.

Show moreHaruki Murakami's Norwegian Wood (1987) veers from his favored detective-fiction genre by offering readers a 1960s coming-of-age romance, a story whose plot nonetheless spins around the protagonist seeking out his personal identity. The conflicts between Japanese tradition and modern, global perspectives are illustrated through the inclusion of popular culture elements such as music, literature and films. This thesis seeks to show how the novel's references to popular culture of the 1960s combine to help the protagonist establish an identity for himself as well as his place within the universal community. First, though, the project explores the impact of the translatability issues that arise with each of the novel's two English translations, variations dictated by the needs of differing audiences. The introduction provides an overview of the study, as well as historical background pertinent to the understanding of the Sixties-era popular culture iconography privileged by Murakami. My methodology favors a cultural studies approach and utilizes reader response and reception theories. Separate chapters then compare specifics between the two translations and examine the functionality and significance of music, literature and film within the novel. The conclusion justifies the subsequent deviations between the translations and argues for the necessity and value of both English versions, but claims Rubin's as the definitive English translation. Likewise, the study of the novel's many popular culture references exemplifies the roles that music, books, and film play in the creation of the protagonist's individual identity in Norwegian Wood while simultaneously illustrating the effectiveness of using globally recognizable media as a bridge between cultures.

Show moreThroughout history, traditional plant breeding has been used to provide resistance to pests, disease and other forms of environmental stress, as well as to increase yield and improve upon quality and processing attributes. Over the last decade, the advancement in sequencing technology and bioinformatic analysis has unleashed a wealth of knowledge about chloroplast genetic organization and evolution. The lack of complete plastid genome sequences is one of the major limitations in advancing plastid genetic engineering to other useful crops. This is due to the fact that plastid genome sequences are essential for the identification of endogenous regulatory sequences and optimal sites for homologous recombination. Analysis of four Solanaceae genomes revealed significant genetic modifications in both coding and non-coding regions. Repeat analysis with Reputer revealed 33 to 45 direct and inverted repeats ? 30bp with at least 90% homology. All but five of the 42 repeats shared among all four genomes were located in the exact same genes or intergenic regions, suggesting a functional role. Intergenic analysis found four regions that are 100 percent identical in all four Solanaceae genomes. Such highly conserved intergenic regions are ideal targets for multi-species transformation cassettes. Protein disulfide isomerases (PDI) are a family of proteins known to function as molecular chaperones and aid in the formation of disulfide bonds during protein folding. They contain at least one thioredoxin domain used for the formation, isomerization, and reduction/oxidation of disulfide bonds. Bioinformatic analysis identified 13 PDI-like (PDIL) proteins found in Arabidopsis that contain at least one thioredoxin domain. In addition to the above-mentioned characteristics, PDIs have been shown to be directly involved in the translational regulation of the psbA mRNA in response to light and could potentially increase the efficiency of chloroplast engineering in plants. Human serum albumin (HSA) requires 17 disulfide bonds to be properly folded and is an ideal candidate for assessing the disulfide bond formation, protein folding, and other chaperone-like characteristics of PDIL proteins. Therefore, I have coexpressed HSA in order to further characterize an Arabidopsis PDIL protein, atPDIL5-4, and in particular, the redox control of the psbA 5'UTR. Interestingly, the polyclonal antibody used for identifying the PDIL protein cross-reacted and identified other proteins, but not the transgenic atPDIL5-4. Results of these investigations will be presented.

Show moreData storage is one of the important and often critical parts of the computing systemin terms of performance, cost, reliability, and energy.Numerous new memory technologies,such as NAND flash, phase change memory (PCM), magnetic RAM (STT-RAM) and Memristor,have emerged recently.Many of them have already entered the production system.Traditional storage optimization and caching algorithms are far from optimalbecause storage I/Os do not show simple locality.To provide optimal storage we need accurate predictions of I/O behavior.However, the workloads are increasingly dynamic and diverse,making the long and short time I/O prediction challenge.Because of the evolution of the storage technologiesand the increasing diversity of workloads,the storage software is becoming more and more complex.For example, Flash Translation Layer (FTL) is added for NAND-flash based Solid State Disks (NAND-SSDs).However, it introduces overhead such as address translation delay and garbage collection costs.There are many recent studies aim to address the overhead.Unfortunately, there is no one-size-fits-all solution due to the variety of workloads.Despite rapidly evolving in storage technologies,the increasing heterogeneity and diversity in machines and workloadscoupled with the continued data explosionexacerbate the gap between computing and storage speeds.In this dissertation, we improve the data storage performance from both top-down and bottom-up approach.First, we will investigate exposing the storage level parallelismso that applications can avoid I/O contentions and workloads skewwhen scheduling the jobs.Second, we will study how architecture aware task scheduling can improve the performance of the application when PCM based NVRAM are equipped.Third, we will develop an I/O correlation aware flash translation layer for NAND-flash based Solid State Disks.Fourth, we will build a DRAM-based correlation aware FTL emulator and study the performance in various filesystems.

The disruption of the steps required for the function of ribosomes represents a potential future target for pharmacological anti-cancer therapy. ABCE1 is an essential Fe-S protein involved in ribosomal function and is vital for protein synthesis and cell survival. Thus, ABCE1 is potentially a great therapeutic target for cancer treatment. Previously, cell biological, genetic, and structural studies uncovered the general importance of ABCE1, although the exact function of the Fe-S clusters was previously unclear, only a simple structural role was suggested. Additionally, due to the essential nature of ABCE1, its function in ribosome biogenesis, ribosome recycling, and the presence of Fe-S within ABCE1, the protein has been hypothesized to be a target for oxidative degradation by ROS and critically impact cellular function. In an effort to better understand the function of ABCE1 and its associated Fe-S cofactors, the goal of this research was to achieve a better biochemical understanding of the Fe-S clusters of ABCE1. The kinetics of the ATPase activity for the Pyrococcus abyssi ABCE1 (PabABCE1) was studied using both apo- (without reconstituted Fe-S clusters) and holo- (with full complement of Fe-S clusters reconstituted post-purification) forms, and is shown to be jointly regulated by the status of Fe-S clusters and Mg2+. Typically, ATPases require Mg2+, as is true for PabABCE1, but Mg2+ also acts as a unusual negative allosteric effector that modulates ATP affinity of PabABCE1. Comparative kinetic analysis of Mg2+ inhibition shows differences in the degree of allosteric regulation between the apo- and holo-PabABCE1 where the apparent Km for ATP of apo-PabABCE1 increases (>)30 fold from ~30 (&)#181;M to over 1 mM when in the presence of physiologically relevant concentrations of Mg2+. This effect would significantly convert the ATPase activity of PabABCE1 from being independent of cellular energy charge to being dependent on energy charge with cellular [Mg2+]. The effect of ROS on the Fe-S clusters within ABCE1 from Saccharomyces cerevisiae was studied by in vivo 55Fe labeling. A dose and time dependent depletion of ABCE1 bound 55Fe after exposure to H2O2 was discovered, suggesting the progressive degradation of Fe-S clusters under oxidative stress conditions. Furthermore, our experiments show growth recovery, upon removal of the H2O2, reaching a growth rate close to that of untreated cells after ~8 hrs. Additionally, a corresponding increase (~88% recovery) in the ABCE1 bound 55Fe (Fe-S) was demonstrated. Observations presented in this work demonstrate that the majority of growth inhibition, induced by oxidative stress, can be explained by a comparable decrease in ABCE1 bound 55Fe and likely loss of ABCE1 activity that is necessary for normal ribosomal activity. The regulatory roles of the Fe-S clusters with ABCE1 provide the cell a way to modulate the activity of ABCE1 and effectively regulate translation based on both cellular energy charge and the redox state of the cell. Intricate overlapping effects by both [Mg2+] and the status of Fe-S clusters regulate ABCE1's ATPase activity and suggest a regulatory mechanism, where under oxidative stress conditions, the translational activity of ABCE1 can be inhibited by oxidative degradation of the Fe-S clusters. These findings uncover the regulatory function of the Fe-S clusters with ABCE1, providing important clues needed for the development of pharmacological agents toward ABCE1 targeted anti-cancer therapy.

Results show that applications' performance varies according to the mapping scheme used, and a dynamic mapping scheme achieves up to 2x increase in peak bandwidth utilization and around 30% higher energy efficiency than a system using only a single fixed scheme Moreover, the technique can be used to limit memory accesses into a subset of the memory devices by controlling data allocation at a finer granularity, providing a method to throttle main memory by allowing un-accessed devices to be put into power-down mode, hence saving power to meet a certain power budget.

Testing of novel compounds includes a preclinical phase where the potential therapeutic is tested in vitro and/or in animal models in vivo to predict its potential efficacy and/or toxicity in humans. The failure of preclinical models to accurately predict human drug responses can lead to potentially dangerous compounds being administered to humans, or potentially beneficial compounds being kept in development abeyance. Moreover, inappropriate choice in model organism for studying disease states may result in pushing forward inappropriate drug targets and/or compounds and wasting valuable time and resources in producing much-needed medications. In this dissertation, models for basic science research and drug testing are investigated with the intention of improving current preclinical models in order to drive drugs to market faster and more efficiently. We found that embryonic rat hippocampal neurons, commonly used to study neurodegenerative disease mechanisms in vitro, take 3-4 weeks to achieve similar, critical ion-channel expression profiles as seen in adult rat hippocampal cultures. We also characterized a newly-available commercial cell line of human induced pluripotent stem cell-derived neurons for their applicability in long-term studies, and used them to develop a more pathologically relevant model of early Alzheimer's Disease in vitro. Finally, we attempted to create an engineered, layered neural network of human neurons to study drug responses and synaptic mechanisms. Utilization of the results and methods described herein will help push forward the development of better model systems for translation of laboratory research to successful clinical human drug trials.

Two main themes were developed as a result of this analysis: the importance of adapting content to French audiences, and the cultural differences between French and American information needs and communication styles. Survey findings were combined with theoretical and practical literature to offer American technical communicators guidance for successful intercultural interactions. This thesis concludes with suggestions for future practice and research in intercultural technical communication.

Show moreHistorically, the United States (US) electric grid has been a stable one-way power delivery infrastructure that supplies centrally-generated electricity to its predictably consuming demand. However, the US electric grid is now undergoing a huge transformation from a simple and static system to a complex and dynamic network, which is starting to interconnect intermittent distributed energy resources (DERs), portable electric vehicles (EVs), and load-altering home automation devices, that create bidirectional power flow or stochastic load behavior. In order for this grid of the future to effectively embrace the high penetration of these disruptive and fast-responding digital technologies without compromising its safety, reliability, and affordability, plug-and-play interoperability within the field area network must be enabled between operational technology (OT), information technology (IT), and telecommunication assets in order to seamlessly and securely integrate into the electric utility's operations and planning systems in a modular, flexible, and scalable fashion. This research proposes a potential approach to simplifying the translation and contextualization of operational data on the electric grid without being routed to the utility datacenter for a control decision. This methodology integrates modern software technology from other industries, along with utility industry-standard semantic models, to overcome information siloes and enable interoperability. By leveraging industrial engineering tools, a framework is also developed to help devise a reference architecture and use-case application process that is applied and validated at a US electric utility.
